| Objective:1.By observing the effects of the three roots balanced acupuncture of Mongolian medicine on exploratory behavior,learning and memory ability and anhedonia level of depressed model rats,it is proved that the three roots balanced acupuncture of Mongolian medicine have significant curative effect on depression.2.By observing the change of the ratio of BDNF/proBDNF and the expression of SERT protein in the raphe nucleus of rats in each group,the mechanism of the treatment of depression by the three roots balanced acupuncture of Mongolian medicine was discussed.Methods:After seven days of adaptive feeding,we randomly divided 24 rats into normal group,model group,drug group and three roots balanced acupuncture of Mongolian medicine group,with 6 rats in each group.The rats in the normal group were raised in a large cage and fed a normal diet without any stimulation.The other three groups of rats were all isolated.The model group was given 8 kinds of stress stimulation for 28 days,and the chronic and unpredictable mild stress model of rat depression was established.The drug group received the same stress stimulation as the model group for 28 days,and after receiving stimulation for1 hour every day,Paluoxiting was prepared into 1mg/m L of purified water and administered intragastric according to 1.8mg/kg body weight.Rats in the three roots balanced acupuncture of Mongolian medicine group received the same stress stimulation as the model group for 28 days,and after receiving the stimulation every day for 1 hour,they were given three roots balanced acupuncture of Mongolian medicine intervention.Intervention methods: Select "Badagan point","Heart point","Heibaiji point" every day,using disposable sterile needle acupuncture.Among them,on the first day of acupuncture intervention,the handle of "Badagan point" was connected to MLZ-Ⅲ intelligent acupuncture treatment instrument of Mongolian medicine,and the temperature was(40±2)℃ for 15 min,while the other two points were not warmed.On the second day,the "Heart point" was warmed by the same method as above,while the other two points were not warmed.On the third day the same method to give the "Heibaiji point" heating stimulation,the other two points do not heat stimulation.Cyclic stimulation for 28 days to prevent burning of the acupuncture site.Within24 hours after 28 days,the modeling was proved to be successful by weight test,sugar water consumption experiment,Open-Field experiment and Morris water maze experiment.Then,the head was severed and the raphe nucleus was taken,and the protein expression of BDNF,proBDNF,Trk B and SERT in raphe nucleus was detected by Western-blot technique,and the ratio of BDNF/proBDNF was calculated.The m RNA protein level of BDNF in raphe nucleus was detected by Real-time PCR.Results:1.Results of weight detection and behavioral experiment: before stress stimulation,there were no significant differences in the results of weight detection,sugar water consumption experiment,OF open box experiment and Morris water maze experiment among all groups(P>0.05).After 28 days of chronic stress stimulation,compared with the normal group,the body weight increase,sugar water consumption and the score of open box experiment of rats in the model group were significantly decreased(P<0.05),and the time to find the platform in the water maze experiment was longer(P<0.05).Compared with model group,body weight increase,sugar water consumption and scores OF open box experiment of rats in drug group and three roots balanced acupuncture of Mongolian medicine group were significantly increased(P<0.05),and the time to find the platform in water maze experiment was shorter(P<0.05).2.The results of Western-blot suggest that the expression level of BDNF protein in the raphe nucleus of rats in the model group was significantly decreased(P<0.05),the expression level of proBDNF and SERT were significantly increased(P<0.05),and the ratio of BDNF/proBDNF was significantly decreased compared with the normal group(P<0.05).The protein levels of Trk B receptors in the raphe nucleus were no prominent differences among all groups(P>0.05).Compared with model group rats,drug group and three roots balanced acupuncture of Mongolian medicine group rats the relative expression of BDNF protein in raphe nucleus increased significantly(P<0.05),relative expression of proBDNF and SERT proteins were significantly decreased(P<0.05),BDNF/proBDNF ratio increased significantly(P<0.05).3.The results of PCR suggest that compared with normal group,the expression level of BDNF m RNA in the raphe nucleus of rats in the model group was significantly decreased(P<0.05).The expression level of BDNF m RNA in the raphe nucleus of rats in three roots balanced acupuncture of Mongolian medicine group and drug group compared with model group was significantly raised(P<0.05).Conclusion:1.The body weight detection and behavioral experiments’ results suggest that the body weight,number of horizontal and vertical movements of open box experiment,sugar water consumption and platform searching time of rats in the three roots balanced acupuncture of Mongolian medicine group were significantly raised.These results revealed that the three roots balanced acupuncture of Mongolian medicine could significantly improve depressed model rats’ depressive state,significantly improve the learning and memory abilities,autonomous activity abilities,spatial exploration abilities and reduce the level of pleasure loss in depression model rats.2.The three roots balanced acupuncture of Mongolian medicine increased the content of BDNF m RNA protein,the ratio of BDNF/proBDNF and decreased the relative expression of SERT protein in the raphe nucleus of depressed model rats.These results suggest that the antidepressant effect of three roots balanced acupuncture of Mongolian medicine may be achieved by increasing the BDNF/proBDNF ratio in the raphe nucleus region and reducing the expression of SERT protein.3.Three roots balanced acupuncture of Mongolian medicine can improve the anhedonia level,learning and memory ability,autonomous activity ability and space exploration ability of depressed model rats,and the mechanism may be related to the increase of BDNF/proBDNF ratio and the decrease of SERT protein expression level in the raphe nucleus. |
