| Objective A depression model of Chronic Unpredictable Mild Stress was established.The expressions of micro RNA-16(mi RNA-16),mature brain-derived neurotrophic factor(BDNF)and probrain-derived neurotrophic factor(pro BDNF)in the raphe nucleus(MRN)of depression rats were detected.To investigate the effect of mi RNA-16 on m BDNF and tropomyosin receptor kinase B(Trk B)signaling pathway.The relationship between BDNF and pro BDNF was determined in the raphe nucleus of depressed rats.It also provides a theoretical basis for the mechanism of acupuncture in the treatment of depression based on the three-acupoints balance theory of Mongolian medicine.Methods Thirty SD rats with the same baseline were randomly divided into five group: a control,a model,a CUMS+drug,a CUMS+acupuncture and a CUMS+anti-mi RNA-16+acupuncture,with 6 rats in each group.The depression model was established by daily chronic unpredictable mild stress.Six rats in the normal group were housed in groups with normal diet and water intake.The rats in the model group were randomly exposed to unpredictable chronic stress for 21 days and then received no treatment.The rats in the CUMS+drug received daily stress stimulation in the same manner as the model group.After 1hour of stimulation,paroxetine was injected via tail vein once a day for 21 days injected dose:2mg/kg and 1mg/ml diluted with normal saline.The rats in the CUMS+acupuncture received the same stimulation method as the model group once a day for 21 days.After 1 hour of stimulation,the rats in the CUMS+acupuncture were given acupuncture intervention,once a day for 21 days.The CUMS+anti-mi RNA-16+acupuncture group received acupuncture combined with tail vein injection of anti-mi RNA-16 once a week for 21 days,1 hour after daily stress stimulation.The behavioral changes of rats in each group were observed before and after the experiment.The expressions of m BDNF,pro BDNF and Trk B in the MRN were detected by Western blot,and the expressions of mi RNA-16 m RNA and BDNF m RNA in the MRN were detected by real-time PCR.Results(1)Before the experiment,There was no obvious abnormality in the behavioral test results of rats in each group before the experiment(P>0.05).After the experiment,compared with the normal group,the model group had reduced horizontal movement,vertical movement and sucrose consumption(P<0.05).Compared with the model group,the CUMS+drug group,CUMS+acupuncture group and CUMS+anti-mi RNA-16+acupuncture group had increased horizontal movement,vertical movement and sucrose consumption(P<0.05).(2)The expression level of mi RNA-16 in the raphe nucleus of the model group was higher than that of the normal group(P<0.05),and the expression of m BDNF m RNA was decreased(P<0.05).The expression of mi RNA-16 m RNA in the MRN was decreased(P<0.05),while the expression of m BDNF m RNA was increased in the CUMS+medication group,CUMS+acupuncture group and CUMS+anti-mi RNA-16+acupuncture group(P<0.05).(3)The expression levels of m BDNF and Trk B in the raphe nucleus of the model group were significantly lower than those of the normal group(P<0.05),the expression level of pro BDNF was increased(P<0.05),and the ratio of m BDNF/pro BDNF was decreased(P<0.05).The relative expression levels of m BDNF and Trk B in the raphe nucleus of rats in the CUMS+drug group,CUMS+acupuncture group and CUMS+anti-mi RNA-16+acupuncture group were significantly higher than those in the model group(P<0.05)and significant reductions in the relative expression of pro BDNF(P<0.05).The ratio of m BDNF/pro BDNF in the MRN was increased in CUMS+medication group and CUMS+acupuncture group(P<0.05).Conclusion(1)Chronic unpredictable mild stress may induce the high expression of mi RNA-16,increase the expression of pro BDNF,and reduce the expression of m BDNF and Trk B in the raphe nucleus of depression rats.(2)Acupuncture of Mongolian medicine may down-regulate the expression of mi RNA-16 and further regulate the m BDNF-Trk B signaling pathway to play an antidepressant role.(3)Chronic unpredictable mild stress can down-regulate the ratio of m BDNF/pro BDNF.The antidepressant mechanism of acupuncture based on the three-root balance theory of Mongolian medicine may be related to the up-regulation of m BDNF/pro BDNF. |
