Objective:Hepatocellular carcinoma(HCC)is the main type of primary liver cancer,which is highly malignant and lacks effective treatment in the advanced stage.The occurrence,development,invasion,and metastasis of HCC are a long-term complex and continuous process of multi-factor interaction.The study of its specific molecular mechanism will help to improve our ability to diagnose and treat HCC.Studies have shown that abnormally high expression of EBNA1-binding protein 2(EBP2)is associated with tumors,but there is insufficient evidence in the field of liver cancer.Therefore,this article aims to explore the expression of EBP2 in HCC,and to explore its impact on the malignant biological phenotype of liver cancer cells.Methods:1.The HCC data set was obtained through the TCGA database,and online databases such as GEPIA,UALCAN,and Kaplan-Meier Plotter were used to analyze the expression difference of EBP2 in HCC and its relationship with the clinical characteristics and survival prognosis of patients.2.Through the study of HPA database and clinical samples of liver cancer,the expression difference of EBP2 protein between HCC cancer tissue and adjacent normal liver tissue was obtained.3.Using Western Blot and q RT-PCR to screen HCC cell lines with high EBP2 expression,and construct EBP2 knockdown cell models.4.The effects of EBP2 on the proliferation,migration and invasion of HCC cells were detected by CCK8 assay,plate colony formation assay,scratch assay and Transwell assay.Results:1.A total of 371 cases of HCC cancer tissues and 50 cases of adjacent normal tissues were obtained from the TCGA database.The analysis results showed that EBP2 was highly expressed in HCC cancer tissues and was related to the patient’s gender and tumor stage.Survival analysis results showed that high expression of EBP2 predicted poor prognosis.2.The immunohistochemical staining images of HCC cancer tissues and adjacent normal tissues were downloaded from the HPA database,and Western Blot was used to detect the expression level of EBP2 in clinical samples of hepatocellular carcinoma.The results showed that EBP2 was significantly overexpressed in HCC cancer tissues.3.At the cell level,two cell lines(Bel7404 and SMCC7721)with relatively high expression of EBP2 were screened out by detecting the protein and m RNA expression of EBP2 in six HCC cell lines.The cell model of EBP2 knockdown was constructed by using small interfering RNA(si RNA),and the successful construction of the EBP2 knockdown model was verified by q RT-PCR and Western Blot experiments.4.Both CCK8 assay and plate colony formation assay showed that the knockdown of EBP2 inhibited the proliferation of HCC cells.Both scratch assay and Transwell assay showed that the knockdown of EBP2 inhibited the migration and invasion of HCC cells.Conclusions:1.EBP2 is highly expressed in HCC and is associated with clinicopathological features and poor survival prognosis of patients.2.EBP2 plays a cancer-promoting role in HCC by affecting cell proliferation and promoting migration and invasion of tumor cell.
