Objective:The benign prostatic hyperplasia(BPH)is a common chronic progressive disease in middle-aged and elderly men,its pathogenesis and development have been widely studied.Aimed to investigate the possible molecular pathological mechanism associated with the clinical progression of benign prostatic hyperplasia(BPH),this study observed the expressions of HIF-1a and CYR61 in symptomatic BPH tissue and normal prostate tissue and analyzed the correlation between them in the clinical progression of BPH.Methods:All tissue specimens were obtained from 10 normal men and 60 BPH cases who underwent transurethral resection of the prostate(TURP)processed and transferred to tissue microarrays.There were used to detect the expression of HIF-1a and CYR61 by immunohistochemical staining,and the correlation between the two in hyperplasia prostatic tissue was analyzed by spearman rank correlation Coefficients.Results:Immunohistochemical staining showed no positive expression of HIF-1a in epithelium and interstitium of normal prostate tissue,whereas CYR61 protein was positively expressed in epithelial cells.In BPH tissue samples,the positive expression rate of HIF-1a was 71.7%(43/60),of which 20 cases(33.3%)were mildly positive(+)and 23 cases(38.3%)were moderately positive(++),with staining mainly located in glandular epithelial cells.The positive expression rate of CYR61 was 65.0%(39/60),of which 20 cases(33.3%)were mildly positive(+)and 19 cases(31.7%)were moderately positive(++),with staining mainly located in the cytoplasm of prostate epithelial and mesenchymal cells.The expression levels of HIF-1a and CYR61 in BPH tissue were significantly higher than in the normal group,and the expression level of CYR61 was positively correlated with HIF-1a(r=0.790,P=0.000).Conclusion:1.While HIF-1 a is not expressed in normal prostate tissue,CYR61 is expressed in epithelial cells of normal prostate tissue.Both are expressed at high levels in the hyperplastic prostate tissue of patients with BPH.2.The expression level of CYR61 was positively correlated with the expression level of HIF-1 a in benign prostate hyperplasia tissue,suggesting that HIF-1 a may have a role in the hyperplasia of prostate tissue in patients through the induction of an increase in CYR61 protein expression.
