Objective: To investigate the association between the potential SNPs rs217727,rs2240183,and rs1333049 of lncRNA and the occurrence of ischemic stroke,and further explore the association between these polymorphisms and risk factors of ischemic stroke.Method: This study selected a population from of Guilin,Guangxi as the research object,including 110 patients with ischemic stroke and 110 healthy controls during the same period.The control group and the case group were matched in age and gender,the clinical data of patients were collected,and DNA was extracted from blood samples of the sample population.Using the method of Sanger sequencing techniques,the samples were genotyped based on the potential functional SNPs rs217727,rs2240183,and rs1333049 that have been screened for ischemia stroke related lncRNA regions.Associated allele and genotype frequencies were analyzed and contrasted using SPSS v26.0.Result:(1)There are genetic polymorphisms in the latent SNP loci rs217727,rs2240183,and rs1333049 of lncRNA in both the case group and the control group.(2)No statistical differences were found between genders for the lncRNA rs217727,rs2240183,and rs1333049 polymorphisms between cases and controls(P>0.05).(3)The rs1333049 polymorphism is associated with susceptibility to ischemic stroke(P<0.05).The CC genotype of rs1333049 confers a higher risk than GG genotype carriers(OR = 4.39,95% CI = 1.51-12.76);In addition,the CC genotype carriers had a higher risk than the GG and GC genotype carriers in the recessive model(OR = 4.6,95% CI = 1.82-11.66),And a 0.94-fold increased risk for each 1 SD increase in the number of C carriers in an additive model(OR =1.94,95% CI = 1.18-3.2).However,rs217727,rs2240183 were not statistically significantly associated with ischemic stroke susceptibility.(4)There was no association between the rs1333049 polymorphism and any of these confounders including age,gender,hypertension,diabetes,or smoking(P>0.05).(5)There was no association between the rs1333049 polymorphism and lipid levels in patients with ischemic stroke(P>0.05).Conclusion:(1)Genetic polymorphisms at the lncRNA rs217727,rs2240183,and rs1333049 loci were identified,and the polymorphisms were not statistically different between genders.(2)Polymorphisms at rs1333049 are associated with susceptibility to ischemic stroke,and the risk of ischemic stroke is increased by the rs1333049 CC gene,recessive model,and an increased number of C alleles.However,rs2240183 and rs217727 were not associated with the risk of ischemic stroke.
