| Objective: To investigate the expression of Keap1 protein and m RN A in the cancerous and paracancerous tissues of patients with stage Ⅰ lun g adenocarcinoma.The clinical data of patients with stage Ⅰ lung adenoca rcinoma and the expression of Keap1 were analyzed to further explore the clinical significance of Keap1 expression in the development of stage Ⅰ l ung adenocarcinoma.Methods: The cancerous and paracancerous tissues of 60 patients wit h stage Ⅰ lung adenocarcinoma who underwent lung cancer resection were retrospectively included.All patients did not receive anti tumor treatment including radiotherapy and chemotherapy before surgery.Immunohistochem ical method was used to detect the expression of Keap1 protein in cancer tissues and adjacent tissues;Quantitative real-time PCR(q RT-PCR)was used to detect the expression of Keap1 m RNA in cancer tissues and adja cent tissues.The experimental results were statistically analyzed based on clinical and pathological data.All data were analyzed using SP-SS24.0 sta tistical software.Results: Results:1.The positive rate of Keap1 protein expression was41.67% in stage Ⅰ lung adenocarcinoma tissue and 10.00% in paired parac ancer tissue.The expression level of Keap1 protein expression in stage Ⅰ lung adenocarcinoma tissue was significantly higher than that in paracance r tissue,and the difference was statistically significant(P<0.05).2.m RNA expression level of Keap1 in stage Ⅰ lung adenocarcinoma ti ssues was 2.79+1.46;The m RNA expression levels in paracancer tissues were 1.00±0.25.The m RNA expression level of Keap1 in stage Ⅰ lung ad enocarcinoma tissues was significantly higher than that in paracancer tissu es,with statistical significance(P<0.05).3.Subgroup analysis showed that the expression rate of Keap1 protein in cancer tissues of patients with stage ⅠA was 31.37%;The expression rate of Keap1 protein in cancer tissue of patients with stage ⅠB was 100.00%,with a statistically significant difference(P<0.05).The m RNA expre ssion in cancer tissue of patients with stage ⅠA was 2.42 ± 1.19,and that of patients with stage ⅠB was 4.89 ± 0.95,with a statistically significant difference(P<0.05).The expression rate of Keap1 protein in cancer tissue of patients with tumor size>3.0 cm was 82.61%;The expression rate of Keap1 protein in cancer tissue of patients with tumor size≤3.0 cm was 16.22%,with a statistically significant difference(P<0.05).The m RNA expr ession level in cancer tissue of patients with tumor size>3.0 cm was 3.93± 1.22;The m RNA expression in cancer tissue of patients with tumor siz e≤3.0 cm was 2.08 ± 1.10,with a statistically significant difference(P<0.05).However,there were no significant differences in protein or m RNA e xpression levels among patients with different age,gender,smoking histor y,and family history(P>0.05).Conclusion: 1.The expression of Keap1 protein and m RNA is incr eased in stage Ⅰ lung adenocarcinoma tissue.2.Expressions of Keap1 protein and Keap1 m RNA showed statisticall y significant differences in TNM stage and tumor size in stage Ⅰ lung ad enocarcinoma patients,but were not related to age,gender,smoking histor y,family history,etc.High expression of Keap1 is associated with the de velopment of stage Ⅰ lung adenocarcinoma,and may become a potential t umor marker and therapeutic target. |
