| Background and purposeAt present,the underlying pathogenesis of Type 2 Diabetes Mellitus(T2DM)is still unknown,and obesity is considered to be one of the main risk factors for the disease.This experiment examined the expression of Proopiomelanocortin(POMC)in the serum of obese patients and T2 DM patients,and further explored the correlation between POMC on obesity and T2 DM.MethodsIn this experiment,four groups of subjects were selected,from April 2019 to January 2021,100 healthy persons and inpatients in the Department of Endocrinology who were examined at the Second Affiliated Hospital of Harbin Medical University.NC group is healthy physical examination(N =24 cases,age 34.71±8.90 years old);Ob group was a simple obese person(N=25 cases,age 38.64 ± 11.88 years old);T2DM group was a pure T2 DM patient(N=26 cases,age 46.85±9.59 years old);The DMOb group was obese patients with type 2 diabetes(N=25 cases,age 40.72±11.24 years).Take the above 4 groups of peripheral venous blood,detecting POMC level used Enzyme-linked immunosorbent assay(ELISA),and record each candidate ’s basic data and clinical testing indicators,and finally compare and correlate the data.Results(1)The expression of POMC in the four groups was statistically significant(P<0.05),the expression level of POMC in the DMOb group(206.25 ±40.38pg/ml)was lower than that of the healthy controls group,Ob group and T2 DM group(P<0.05,509.13±109.01pg/ml,350.54±66.64pg/ml,255.59±50.80pg/ml).And the expression level of POMC in T2 DM group was lower than that in Ob group and NC group(P < 0.01).POMC level in Ob group was lower than that in NC group(P <0.01).(2)The correlation analysis results between POMC expression level and various indicators show: POMC expression level and BMI,WHR,waist circumference,hip circumference,HbA1 c,FPG,GA,FINS,HOMA-IR,TG,LDL-C levels were negatively correlated(P<0.05,r =-0.329,-0.541,-0.615,-0.555,-0.787,-0.699,-0.572,-0.423,-0.613,-0.421,-0.307).POMC expression level and HDL-C levels were positively correlated(P<0.001,r = 0.402).Age,gender and TC are not related to POMC level(P> 0.05,r =-0.194,-0.168,-0.145).Line partial correlation analysis showed that the expression level of POMC was still highly correlated with BMI,waist circumference,hip circumference,WHR,HOMA-IR,HbA1 c,GA,FINS,FPG,TG,LDL-C,HDL-C(P<0.05,r=-0.387,-0.733,-0.632,-0.663,-0.557,-0.67,-0.393,-0.364,-0.652,-0.396,-0.318,0.481).(3)Multiple linear regression analysis was conducted with BMI,HbA1 c,LDL-C as independent variables and POMC expression as dependent variable.The results showed that HbA1 c is statistically significant(P<0.001),and negatively correlated(b’=-0.568).ConclusionsThe expression of POMC in the serum of patients with type 2 diabetes and obesity decreased significantly,and the content of serum POMC in patients with type2 diabetes was lower than that in patients with simple obesity.And the expression level of POMC is negatively correlated with BMI,TG,HbA1 c,HOMA-IR;and there is a linear regression relationship between the expression level of POMC and HbA1 c.These results suggest that POMC may be related to the occurrence of obesity and type2 diabetes. |
