| Objective:The modern biological basis of ischemic stroke with Qi deficiency and blood stasis syndrome(IS-XY)will be clarified by the regulation of iron transport network point and positive and negative aspects.The research results provide an important basis for further understanding of the essence of IS-XY,and it is of great significance to promote the clinical application of Yi Qi Huoxue principle in the prevention and treatment of ischemic stoke.Methods:Diseases and syndromes combined with modeling methods to replicate the IS-XY model.In the first part of the experiment,7-9 week old SD male rats were randomly divided into sham group,model group,Buyang Huanwu Decoction group,and Xuefu Zhuyu Decoction group(divided into 1 day,3 days,and 7 days),the neuroprotective effect of Buyang Huanwu Decoction on rats with IS-XY was observed by the score of neurological deficit signs,TTC staining,HE staining and Nissl staining.The second part of the experiment is grouped the same as the first part of the experiment.DMT1,TfR,Fpn1,hepcidin and ferritin on the cortex and hippocampus are observed by immunofluorescence,Western blot,and qRT-PCR.Results:1.Compared with the model group and the Xuefu Zhuyu Decoction group,Buyang Huanwu Decoction reduced the neurological deficit score and cerebral infarction volume in the rat model of IS-XY(P<0.05).2.Compared with the model group and the Xuefu Zhuyu Decoction group,the changes of cerebral infarction and pathological changes in the cortex and hippocampus of the Buyang Huanwu Decoction group were lighter.3.Compared with the model group and the Xuefu Zhuyu Decoction group,Buyang Huanwu Decoction decrease the protein expression of DMT1,TfR,and hepcidin(P<0.05),and increase the protein expression of Fpn1 and ferritin(P<0.05).4.Compared with the model group and the Xuefu Zhuyu Decoction group,Buyang Huanwu Decoction decrease gene expression of DMT1,TfR,hepcidin(P<0.05),and increase gene expression of Fpn1 and ferritin(P<0.05).Conclusion:1.Buyang Huanwu Decoction and Xuefu Zhuyu Decoction can reduce brain tissue edema and necrosis,reduce neuronal degeneration and death,thereby reducing neurological deficit score and cerebral infarction volume,so as to improve brain damage in rats with IS-XY.And the purpose of promoting the recovery of brain nerve function.2.In the IS-XY model,Buyang Huanwu Decoction)is better than Xuefu Zhuyu Decoction in improving the degree of brain damage and promoting brain nerve function.3.Buyang Huanwu Decoction and Xuefu Zhuyu Decoction inhibit the expression of iron transfer proteins(DMT1,TfR,hepcidin),and promote iron transfer proteins(Fpn1) and cellular iron Storage protein(ferritin)expression in IS-XY.4.In the IS-XY model,Buyang Huanwu Decoction is better than Xuefu Zhuyu Decoction in regulating iron transport function.5.Buyang Huanwu Decoction inhibits the transfer of iron ions into the cells and promotes the elimination of iron ions,reduces the deposition of iron ions in the cells and increases the combination of iron ions and ferritin to form non-toxic complexes to achieve the purpose of regulating iron metabolism. |
