| Objective: To study the model construction of coronary heart disease(CHD)with blood stasis syndrome(BSS)in rats,and construct the formation stages of CHD with blood stasis syndrome(BSS)through high-fat feed combined with vitamin D3 gavage and ISO injection.Study and evaluate the model at multiple levels in terms of macroscopic observation,objective indicators and microcosmic syndromes,so as to explore the establishment and evaluation of the model of combination of diseases and syndromes in traditional Chinese medicine.Methods: Fifty-one male SD rats were randomly divided into normal group(15 rats)and model group(36 rats).Among them,the model group and specific number were 12 rats in the pre-blood stasis syndrome group,12 rats in the sub-blood stasis syndrome group,and 12 rats in the heart blood stasis syndrome group.The rats in the model group were fed with high-fat diet,combined with vitamin D3 and injected with isoproterenol subcutaneously to make the model of coronary heart disease with blood stasis syndrome.During this period,the general state,tongue picture and hind paw picture of rats in each group were collected,the blood lipid and hemorheology levels of rats in each group were measured,the aorta was stained with HE,and the serum TNF was measured by Elisa method-α、IL-6、sICAM-1、sVCAM-1.Finally,SPSS26.0 statistical software is used to analyze the data.Results:(1)Statistical analysis of tongue picture showed that:(1)Compared with the normal group,the R value,G value and B value in the early stage of blood stasis syndrome group increased to different degrees;Compared with the normal group,the G value of the sub-blood stasis syndrome group continued to rise on the basis of the previous stage,but the G value and B value had no statistical difference;Compared with the normal group,the B value of heart blood stasis syndrome group was higher,but lower than that of sub-blood stasis syndrome group.(2)Compared with the early stage group of blood stasis syndrome,the R value,G value and B value of the sub-blood stasis syndrome group and the heart blood stasis syndrome group increased in varying degrees,but the growth rate was slightly slower than the previous stage.(3)Compared with the sub-blood stasis syndrome group,the R value and G value of the heart blood stasis syndrome group increased,but the B value decreased.(2)Through statistical analysis of the pictures of the hind paws,it was found that:(1)Compared with the normal group,the R value,G value and B value of the early stage group of blood stasis syndrome decreased;Compared with the normal group,the R value,G value and B value of the sub-blood stasis syndrome group decreased,and the R value and B value kept decreasing on the basis of the previous period,while the G value increased on the contrary;Compared with the normal group,the R value and B value of heart blood stasis syndrome decreased.(2)Compared with the early stage group of blood stasis syndrome,the R value and B value of the sub-blood stasis syndrome group decreased,while the G value increased;Compared with the pre-blood stasis syndrome group,the R value and B value of the heart blood stasis syndrome group decreased,and the G value was not statistically significant.(3)Compared with the sub-blood stasis syndrome group,the R value,G value and B value of the heart blood stasis syndrome group decreased.(3)The blood lipids of rats in each group were detected,and the results showed that:(1)Compared with the normal group,the levels of TC,HDL-C and LDL-C in the early stage of blood stasis syndrome group increased,in which the levels of TC and LDL-C increased significantly,and the levels of TG decreased;Compared with the normal group,the levels of TC,HDL-C and LDL-C in the sub-blood stasis syndrome group increased,and the levels of TG decreased,which continued to decrease on the basis of the pre-blood stasis syndrome group;Compared with the normal group,the level of TG increased and the level of HDL-C decreased.(2)Compared with the pre-blood stasis syndrome group,TC and LDL-C in the sub-blood stasis syndrome group continued to increase,while TG continued to decrease;Compared with the pre-blood stasis syndrome group,the heart blood stasis syndrome group increased on the basis of the continuous decline of TG in the sub-blood stasis syndrome group.(3)Compared with the sub-blood stasis syndrome group,the TC in the heart blood stasis syndrome group decreased,even lower than the normal group,and the levels of HDL-C and LDL-C were significantly decreased.(4)The blood rheology of rats in each group was tested.The results showed that:(1)compared with the normal group,the plasma viscosity in the early stage of blood stasis syndrome group was higher,and the viscosity in the middle and high shear was significantly higher;Compared with the normal group,the plasma viscosity of the sub-blood stasis syndrome group continued to increase on the basis of the pre-blood stasis syndrome group;Compared with the normal group,the plasma viscosity level in the period of heart-blood stasis syndrome decreased,and the viscosity in the middle and high shear increased.(2)Compared with the pre-blood stasis syndrome group,the plasma viscosity of the sub-blood stasis syndrome group continued to rise,and the viscosity levels of low shear,middle shear and high shear were significantly decreased;Compared with the early stage group of blood stasis syndrome,the viscosity levels of low shear,middle shear and high shear in the stage group of blood stasis syndrome were significantly decreased.(3)Compared with the sub-blood stasis syndrome group,the plasma viscosity of the heart blood stasis syndrome group decreased.(5)Compared with the aortic HE staining,the results showed that the three layers of the intima,media and adventitia of the aorta in the normal group were complete,with clear layers,and the elastic fibers of the media were smooth,with basically the same direction.The HE staining of the aorta of rats in the sub-blood stasis syndrome group showed that the surface of the intima was obviously not smooth,and there was irregular gray plate bulge,which was distributed dispersedly.Because of the unstable plaque falling off,cracks and cavities appear in the membrane.HE staining highlights that the intima and adventitia are damaged to a certain extent.At the same time,it damages the structure of the middle membrane,destroys the elastic fibers,and causes the atrophy of the aortic cells,especially the smooth muscle cells.The HE staining of the aorta of rats in the heart blood stasis syndrome group showed that the surface of the intima was extremely unsmooth,and even ruptured.The surface was scattered with irregular gray plate-like protrusions,and there were complete or incomplete cracks and cavities,indicating that the intima,media and adventitia of the aorta were damaged to a large extent,leading to the destruction of vascular elastic fibers and the serious damage of aortic smooth muscle.(6)The above tests were carried out on rats in each group,and the results showed that:(1)Compared with the normal group,TNF in the early stage of blood stasis syndrome group-α、The levels of IL-6 and sVCAM-1 increased;Compared with the normal group,the level of sVCAM-1 in the sub-blood stasis syndrome group increased;Compared with the normal group,the levels of sICAM-1 and sVCAM-1 in the group of heart blood stasis syndrome increased.(2)Compared with the pre-blood stasis syndrome group,the TNF of the sub-blood stasis syndrome group-α、The levels of IL-6,sICAM-1 and sVCAM-1 increased;Compared with the pre-blood stasis syndrome group,the heart blood stasis syndrome group TNF-α 、 sICAM-1 and sVCAM-1increased.(3)Compared with the sub-blood stasis syndrome group,the heart blood stasis syndrome group has TNF-α、 The levels of IL-6,sICAM-1 and sVCAM-1 increased.Conclusion: The experiment shows that the formation stages of blood stasis syndrome of coronary heart disease are more consistent with the pathogenesis and development of blood stasis syndrome of coronary heart disease in terms of macroscopic observation,objective indicators and microcosmic aspects of syndromes through the combination of high-fat feed and vitamin D3 intragastric infusion and ISO injection,which shows that this model is more consistent with the animal model of blood stasis syndrome of traditional Chinese medicine,and the study of blood stasis syndrome of coronary heart disease in traditional Chinese medicine is more reasonable. |
