Objective: To study the effect of early intervention of Xueshuantong injection combined with rt-PA on the expression of Neu N,GFAP and TNF-α in rats with acute cerebral infarction,and to explore its mechanism on acute cerebral infarction,so as to provide scientific basis for further clinical research.Methods: SD rats were randomly divided into Sham group,MCAO group,rt-PA group and XST group.Each group was subdivided into 1 d,3 d and 7 d subgroups,with 5 rats in each time subgroup.The Sham group and the MCAO group were given intravenous injection of 0.9%sodium chloride solution of the same volume,the rt-PA group was given intravenous injection of alteplase and 0.9% sodium chloride solution of the same volume,and the XST group was given intravenous injection of alteplase and Xueshuantong injection.The death rate of rats in each group was calculated,and the m NSS score was improved at the corresponding time point.The brain was decapitated and the brain was taken.The infarct area was observed by TTC staining,the morphological changes of neurons in the ipsilateral hippocampus were observed by HE staining,and the expression levels of Neu N,GFAP and TNF-α in the ipsilateral hippocampus were detected by IHC method.Results: 1.Mortality: Except for the Sham group,there was no significant difference in mortality among the other groups(p>0.05).2.TTC staining:(1)the cerebral infarction area at each time point in the rt-PA and XST group was significantly smaller than that in the MCAO group(p<0.01);(2)compared with rt-PA group,the area of cerebral infarction at each time point of XST group was significantly smaller than that of thrombolysis group(p<0.01).3.HE staining:(1)The structure of nerve cells in the Sham group was clear,neatly arranged,the nucleus was lightly stained and located in the center of the cells;(2)the number of nerve cells in the ipsilateral hippocampus of the MCAO group decreased,the nucleus pyknosis,deep staining and was squeezed to the edge,part of the cells swelled,the intercellular space widened,degeneration,necrosis and vacuoles occurred;(3)compared with the MCAO group,the vacuolar degeneration,nucleolar shrinkage,degeneration and necrosis,widening of tissue space,glial cell proliferation and pathological morphology of brain tissue were significantly reduced in rt-PA and XST group at each time point.4.The m NSS score:(1)Compared with the Sham,the score at different time points in each group was significantly higher than that in the Sham(p<0.01);(2)the score at 1 day of the rt-PA group was lower than that in the MCAO group(p<0.05)and the scores of the rt-PA group at 3 days,7days and XST group at different time points decreased significantly(p<0.01);(3)compared with the rt-PA group,the score of XST group decreased on the 7th day(p<0.05).5.Neu N:(1)Compared with Sham group,the expression of Neu N in MCAO group,rt-PA group and XST group decreased significantly at each time point(p<0.01);(2)compared with MCAO group,the expression of Neu N in rt-PA group and XST group increased significantly at each time point(p<0.01);(3)compared with rt-PA group,the expression of Neu N expression in XST group at 1 day and 3 days increased(p<0.05)and at 7 days significantly increased(p<0.01).6.GFAP:(1)Compared with Sham,the expression of GFAP in MCAO group,rt-PA group and XST group increased significantly at each time point(p<0.01);(2)compared with MCAO group,the expression of GFAP expression in rt-PA group at 1day and 3 days increased(p<0.05),and at7 days of the rt-PA and at each time of the XST significantly increased(p<0.01);(3)compared with rt-PA group,the expression of GFAP expression in XST group at 1day and 3 days increased(p<0.05)and at 7 days significantly increased(p<0.01).7.TNF-α:(1)Compared with Sham,the expression of TNF-α in MCAO group,rt-PA group and XST group increased significantly at each time point(p<0.01);(2)compared with MCAO group,the expression of TNF-α in rt-PA group and XST group decreased significantly at each time point(p<0.01);(3)compared with rt-PA group,the expression of TNF-α in Xueshuantong group decreased on the 3rd(p<0.05)and decreased significantly on the 7th day(p<0.01).Conclusion: 1.Xueshuantong injection is effective within 24 hours after rt-PA intravenous thrombolysis in MCAO rats,and does not increase the risk of death in rats.2.Xueshuantong injection combined with rt-PA can increase the expression of Neu N,decrease the expression of GFAP and TNF-α,and improve the symptoms of neurological deficit and brain morphology,suggesting that inhibition of neuronal death,astrocyte activation and inflammation may be one of the neuroprotective mechanisms of early intervention of Xueshuantong injection combined with rt-PA on acute cerebral infarction.
