Rhythmic gymnastics has experienced more than 60 years of development,difficult movement is always the main technical line of the whole sport.Under the guidance of artistic gymnastics competitive events and project rules,the types and structures of difficult movements have changed a lot,and the technical characteristics gradually tend to be difficult,accurate and complex,which has a high requirement for the athletes’ technical completion quality and physical quality.Heavy load and high-intensity training have caused great pressure on the ankle joint.Especially,the injury of ankle ligament is more prominent in the difficult movement training,so it is urgent to seek effective and advanced treatment.Stem cells have the potential of multidirectional differentiation.Stem cell transplantation can replace damaged cells in the body and repair tissues.Stem cell therapy has developed rapidly in the field of biomedicine,and even some of them have been applied in clinical experiments in medicine and disease research.Faced with the high rate of sports injury,the trend of stem cell therapy for sports injury has attracted the attention of many researchers and professionals.In this paper,the injury sites and types easily caused by the difficult movements of rhythmic gymnastics were investigated by questionnaire,experiment and literature,etc.,and a mouse ankle ligament injury model with the same pathology was established.Biological detection of target stem cells was conducted and the affected areas of mice were transplanted to explore the effect of mesenchymal stem cells in the treatment of ankle ligament injury.This paper draws the following conclusions:1.Ankle ligament injury rate caused by rhythmic gymnasts in difficult maneuvers: the injury rate of difficult movements is as high as 75%;The most likely injuries are ankles,feet,and knees;The types of ankle injuries are mainly in ligaments,tendons,and muscles,of which ligaments are the most damaged;Ankle ligament injuries are mainly caused by insufficient flexibility,insufficient strength,and decreased control;Rhythmic gymnasts believe that the ankle and ligament tissues are the ankle and ligament tissues that have a greater impact on the injury site and type.2.Treatment status of rhythmic gymnasts after ankle toughness injury caused by twisting difficult movements: 43.75% were mild injuries,31.25% were moderate injuries,and 25% were severe injuries;The main treatment methods are traditional medical therapy and physical therapy.The recovery time is at least one month.It is not easy to recover after ankle ligament injury.3.Bovine lung mesenchymal stem cells were obtained from bovine lung tissue of fetal cattle,and the target stem cells were obtained through tissue separation and adherence.After the cells were gradually purified during the passage process,the cells showed obvious long spindle shape.Secondly,the growth curve was drawn,showing an "S" shape distribution,and the growth rate was P18>P11>P7.The clonogenesis ability test results showed that the clonogenesis ability was related to the cell growth rate,which proved that the proliferation ability of cell culture in vitro was consistent with the dry characteristics of mesenchymal stem cells.After induction differentiation of lipogenesis,osteoblast and chondroblast,lipid cells showed obvious oil droplets,and bone cells and chondrocytes showed corresponding nodules.Relevant cell marker genes were detected by PT-PCR,and the specific genes of lipid cells were PPAR-γ and COL1A2,while those of bone cells were OPN and COL2A1.Chondrocytes showed positive expression of COL2A1,VIM and ACAN.Finally,the surface markers of bovine lung mesenchymal stem cells were detected by immunofluorescence,and CD29,CD44,CD71,CD105 and CD90 were positively expressed,while CD34 was negative and not expressed in the control group.The karyotype confirmed that the stem cells were normal diploids with no gene difference.4.The same pathological mechanism of ankle ligament injury was used to conduct collagenase-induced ankle ligament injury model on mice.CM-DIL tracer labeling was performed on bovine lung mesenchymal stem cells described above,and the affected areas of mice were transplanted.The results showed that there was no difference between the mice in the treatment group and the normal group after stem cell transplantation,indicating that the mouse ankle ligament injury model was successfully prepared,and it was confirmed that stem cells have certain effects on the treatment and repair of ankle ligament tissue.Combined with the above analysis on the prospect of the clinical application of stem cells,and some of the clinical studies on the treatment of human injuries have also achieved good results,and the application of stem cells to the treatment of human sports injuries has opened up a new way.5.Based on the technical requirements of difficult movement,the ankle joint is in a state of high heel for a long time,and the ligament is overstretched.When the joint homeostasis is broken,the sprain will occur.Compared with conventional therapies,stem cell therapy has certain advantages.It mainly repairs inflammatory factors and cells through cells,thereby improving the internal environment and contributing to the perfection of tissue function.Therefore,it is necessary to use the mouse injury model and according to the same pathology of ankle ligament injury caused by difficult movement,to conduct stem cell transplantation and test the data results,so as to confirm the effect of stem cell therapy,promote the clinical application of injury,relieve the pain of difficult movement injury of rhythmic gymnasts and extend their sports life.
