Experiment Of SHENLUONING Which Effect On IgA Nephropathy Model Mouse Mesangial Cell, Extracellular Matrix And Cytokines

Objective: IgA nephropathy(IgAN) is common pathologic type of primary glomerular disease .and it is the main season of 2%~3% patients turning to end stage renal failure. The research to investigate the effects of shenluo-ning(SHLN) on glomerular cytokine,mesangial cell and extracellular matrix on replication IgAN mouse model,discuss the action machanism of the prescraption treating IgAN.Method: Replicate IgAN mouse model by taking orally and time-controlled vein injecting bovine serum albumin(BSA),vein injecting staphylococcus enterotoxin B (SEB),giving SHLN treatment. At the same time shenyan-kangfupian(SHYKFP) act as reference group,to observe blood urine nitrogen (BUN),creatinine (Cr),urine erythrocyte,24 hours urine protein quantity,kidney morphology,ultrastructure and immune complex deposition. Using enzyme linked immunosorbent assay (ELISA), radio-immunity,immunohistochemistry method and computer image analysis, Retrotranscription-Polymerase chain reaction( RT-PCR),to observe the effects of SHLN on IL-1, TNF-ct, IL-lra, IL-10, a-SMA, ICAM-1 and FN of IgAN mouse model tenal tissue. Results: (l)Content of urine erythrocyte, urine protein, BUN and Cr in SHLN group remarkablely decreased comparing with that of in model group (p<0.05), especially more better than SHYKFP group in decreasing urine erythrocyte (p<0.05). (2)Normal group has no IgA and IgG deposition; model group has widespread IgA and diffuse IgG deposition; IgA and IgG deposition intensity of SHLN is less than model group. The same result under the light microscope and electron microscope. (3) The result of PAS stain computer image analysis show that area ratio value of mesangial and glomerulars of model group is higher than normal group (p<0.05). SHLN group and SHYKFP group is better than model group (p<0.05). moreover SHLN group is better than SHYKFP group (p<0.05). (4) Expression of a-SMA, ICAM-1 and FN in glomerular of model group are higher than normal group (p<0.05); SHLNgroup and SHYKFP group is lower than model group (p<0.05). (5) Content of IL-1, TNF-a in kidney tissue of model group is higher than normal group (p<0.05). IL-1, TNF-a of SHLN group is lower than model group remarkablely; moreover, TNF-a content in SHLN group is lower than SHYKFP group (p<0.05). (6)Content of IL-lra, IL-10 of model group is little higher than normal group; the increase of IL-lra content is significant (p<0.05). content of IL-lra, IL-10 of SHLN group and SHYKFP group are all higher than model group (p<0.05), SHLN group is higher than SHYKFP group(p<0.05).Conclusion: (1) SHLN is one of the effective prescraption for IgAN. It can releive the urine erythrocyte,protein content and decrease BUN,Cr of IgAN model mouse. (2) SHLN relieve pathologic lesion of kidney tissue ,inhibite glomerulars a-SMA, ICAM-1 and FN expression.the function probably relate to SHLN can decrease IL-1, TNF-a,increase IL-lra, IL-10,regulate abnormal cytokine net. (3)The multi-target function of SHLN maybe is the machanism how it cure IgAN and prevent the progressive development.
