The Influence On The Experimental Animals Of The Injections Of Compound Medicine To The Epidural Space

Objective: To study whether the physical and chemical characters are stable after each own component of reuniting liquid from three different compound medicine. Further more, each general and local influences to the rabbit after the injection of the three different formulate reuniting liquid into its epidural space are learned from endocrine,pathophysiology, nerve electrophysioloy and histological pathology.Method: 1. The color, specific gravity and pH of each liquid are recorded, and also, the absorption spectrum of each liquid and each components were measured by using UV spectrophotometer. 2. The fasting blood glucose value before and after injection of each group was measured by using glucoxidase test and compared with reference group to study the influence of the injection. Further more, the influence to the Hypothalamus- Pituitary -Adrenal(HPA)axis by injection was suspectedï¼› 3.The content of SOD before and after injection was measured by usingxathine oxidase test. From this way, the chemical injury of spine and nerve by the three reuniting liquid was suspectedï¼›4. Electrophysiological index before and after injection was recorded by using electromyograph so the neural electrophysiological influence by the injection could be observed. 5. Organism pathological exam was done by macrosomia observing, HE staining, specific staining, immunohistochemical staining and LFB staining. Microstructure was also be observed by using transmission electron microscopy(TEM).Result: 1. The SG of the three reuniting liquid was almost as the same as the water. The value of pH was all acidity, the sequence is: B>D>C. After the imbrication and mixture of its components, the absorption spectrum of each reuniting liquid was almost the same with all its compound; 2. The fasting blood Glu value of group B, C and D after injection by operating method was all higher than the value before injection(p<0.01) and significant different with the value of group A(p<0.01]. the value of blood Glu of group A after operation was notable higher than the value before operation. The value of blood Glu was insignificant while 4 weeks after injection(P>0.05); 3. The SOD value of group B, C and D was not notable before and after injection, and was not notable compared with group A(p>0.05), too; 4. The electrophysiological exam of group B, C and D was insignificant before and after injection(p>0.05), and was insignificantcompared with group A(p>0.05), too; 5. The notable proliferation was not observed by eyes in the local of injection. The infiltration of inflammation cells and mild proliferation of fibrocyte around dura mater was observed by HE stained in 4 and 8 weeks after injection. Infiltration and exudation of inflammation cells was observed by HE stained in epidural nerve root. Compared with group A, no changes of group B, C and D were observed under specific stained. Proliferation of type â…¡ collagen fibers around dura mater was seen under immunohistochemical stained in 4 and 8 weeks after injection. There is no significant demyelination changes under LFB stained. The thickness and shape of the myelin sheath in epidural nerve root was not regular under transmission electronic microscopy in 4 and 8 weeks after injection. Fibroblast was also seen there. In nerve endometrium, macrophage could be seen under TEM, myelinated nerve fiber changed significantly, but nonmyelinated nerve fiber changed mildly. When 8 weeks, the changes of group D is smaller than the group B and C.Conclusion: 1.Through pharmacological exam, we found that the pH value of the three reuniting liquid in epidural space was acidity. Every components of each liquid were stable and no chemical reaction was happened; 2. The blood Glu value was notable increased by giving medicines through operation method.Maybe, the operation was the course of the increase. But steroid used in epidural should be considered also.
