Part One: Basic Study1, Expression of Matrix Metalloproteinase and Nitric Oxide Synthase of Pituitary Adenomas and Its Relationship With The Invasion of Pituitary AdenomasObjective: In this research,we detect the expressions of MMP2,MMP9, iNOS, nNOS and microvessel density (MVD) in pituitary adenoma tissues . This will help us to realize the correlation between MMPs(MMP2, MMP9), NOS(iNOS, nNOS) and MVD and its relation with the invasiveness of pituitary adenoma. Materials and Methods: (1) 44 cases specimens of pituitary adenomas , 23 cases of them are invasive pituitary adenoma, 21 cases are non-invasive pituitary adenomas. (2) Using immunohistochemical SP staining, We detected MVD (labeled by CD34), MMP2, MMP9, nNOS, iNOS in paraffin slices of 44 pituitary adenoma tissues. (3) Relationships between MMP2, MMP9, nNOS, iNOS expression and MVD were analyzed in pituitary adenoma tissues, and so were relationships between MMP2, MMP9, nNOS, iNOS expression, MVD and invasiveness of pituitary adenoma. Results:There is no obvious difference in age and sex (p>0. 05).(1) The positive expression rate of MMP2 in non-invasive pituitary adenomas and invasive pituitary adenomas is 10% ,43.5% respectively. There is significant difference between 2 groups (p=0. 017). (2) The positive expression rate of MMP9 in non-invasive pituitary adenomas and invasive pituitary adenomas is 20% ,65.2% respectively. There is significant difference among them(p=0. 026). (3) The positive expression rate of nNOS in non-invasive pituitary adenomas and invasive pituitary adenomas is 0%, 56.5% respectively.There is significant difference between 2 groups (p=0. 002). (4) The positive expression rate of iNOS in non-invasive pituitary adenomas and invasive pituitary adenomas is 20%, 73.9% respectively.There is significant difference between 2 groups(p=0. 007). (5) The mean value of MVD is significant difference between non-invasive pituitary adenomas group(13. 9) and invasive pituitary adenomas group(45. 2)(p=0. 016). There is positive correlation between MMP2, MMP9, nNOS, iNOS expression and MVD. Conclusions : (1) The expression of MMP2,MMP9, iNOS, nNOS in invasive pituitary adenomas is higher than that in non-invasive pituitary adenomas. (2) There is positive correlation between MMP2, MMP9, nNOS, iNOS expression and MVD, which suggest MMP2, MMP9, nNOS, iNOS can facilitate pituitary adenomas growth by accelerating angiogenesis. (3) % MMPs, NOS play an important role in invasiveness of pituitary adenomas. The expressions of them can be potential invasiveness and prognostic markers of pituitary adenomas.
