Endothelin(ET) is the most potent vasoconstrictor peptide with strong mitogenetic effects. Experimental studies have sugggested that it may excert an important role in the pathophysiology of cardiolvascu-lar diseases. This study aims to investigate the changes of ET in several diseased conditions for searching indications of ET antagonists or endothelin converting enzyme inhibitors(ECEl). 1. Plasma ET levels in coronary circulation in patients with CHDSlection of patients: (1)CHD group: 23 patients with more than 50 % stenosis of coronary artery, of which 16 patients( 70 %) had severe stenosis. (2)Control group: 10 subjects with normal coronary artery in angiogram. There are no significant differences in age, blood pressure, blood glucose, plasma lipid and cardiac function between both groups.Method and results: To measure plasma ET levels,blood samples werewithdrawn simutaneously from ascending arota(AO),coronary sinus(CS)andiliac vein (IV) before coronary angiography. The results showed that therewere no significant difference in every site between two groups and alsono differences among AO.CS and IV in the same group(p>0.05). Theseresults indicated that coronary atherosclerosis had no effects on theplasma ET levels.2. Effects of experimental atherosclerosis on the ET contents in the plasma and vascular tissueExperimental design: Model of atherosclerosis in the bilaterl iliac arteries of 6 rabbits were established through denuding vascular endothelum followed by high cholesterol meal (group A). Other 6 rabbits were only fed with high cholesterol meal serving as control (group B). Blood samples in the proximal and distal atherosclerosis in group A and the corresponding sites in group B were withdrawn for the measurement of plasma ET levels.
