A claster of monoclonal antibodies against different monocyte/macrophage subtypes of rabbits were established, named as MH1, MC7, MB9, MB6, MG8 and MF11 respectively. The choromsome number of them were MH1/106, MC7/118, MB9/ 109, MB6/114, MG8/121 and MF11/107, and the IgG subtypes were MHl/IgG3, MC7 and MB9/IgG1, MG8 and MB6/IgG2a, MF11/IgG2b seperately. For these 6 McAbs, there were different reactions with the periphiral blood monocytes and macro-phages harvested from different origin, but no cross-reaction noticed with other blood cells and SMC. The results obtained by flow cytometry analysis showed that the positive rates of periphiral monocytes and macrophages from various origin were significantly different after reacted with these six labelled McAbs separately. Antigens of them on cell membrane were analyzed via western blot method, and the molecular weight involved in them were MH1/ 54KD, ,C7/20KD, MB9/96KD and MF11/37KD respectively.By using of these 6 McAbs, the distribution of different monocyte/macro-phage subtypes in aortic atherosclerotic lesions were investigated through immunohistochemical thchnique. Results showed that the infiltrative mono-cytes/macrophages in aortic lesions were mainly positive for McAb MHl as well as MC7, less cells positive for McAb MB9 or MB6, and only few cells positive for McAb MG8 and MFll.The effect of McAb MHl and MC7 on the development of experimental atherosclerosis in vivo was studied in cholesterol-fed rabbits (New Zealand white rabbits).A dosage of 0.2 g cholesterol/ kg body weight/ day was given to rabbits of the experimental groups for totally 10 weeks.
