Research Of Shenshuaibaoshen Capsule On Delaying Chronic Renal Failure And Effecting TGF-β1/Smad Signal Transduction Pathway

Objective: To investigate the effect of the Shenshuaibaoshen Capsule to kidney function of adenine caused chronic renal failure rats and TGF-β1/Smad signal transduction pathway, and to illustrate the mechanism of Shenshuaibaoshen Capsule on delaying chronic renal failure and relieving renal interstitial fibroblast. Methods: In the experiments, the adenine caused chorinic renal failure model is adopted. It is observed for the change of the common physical order, urine quantity, 24 hours uPro, BUN and Scr in blood serum of experimental rats. HE and Masson staining method for pathologic slice are used to observe the pathlology change of the experimental rats kidney tissue afer therapied and the phase of renal fibroblast. The protein expression of TGF-β1, Smad2/3, Smad7 in kidney tissue are detected respectively by immunohistochemistry with two steps. The expression of TGF-β1 receptor-ⅠmRNA and TGF-β1 receptor-ⅡmRNA are also detected by semi-quantitative RT-PCR. Results: The physical status and sympotum of the CRF rats are improved obviously after treating by Shenshuaibaoshen Capsule, and the 24h uPro is reduced which has the significant difference compared with antithesis groups. The Shenshuaibaoshen Capsule reduces the expression of TGF-β1 protein in kidney tissue, adjusts the Smad2/3 to low expression and the Smad7 to high expression, which has the significant difference compared with antithesis groups. The expression of TGF-β1RⅠ,ⅡmRNA are also reduced which have significant difference with antithesis groups. As a result, the TGF-β1/Smad signal transduction pathway is blocked by the Shenshuaibaoshen Capsule. Conclusions: The Shenshuaibaoshen Capsule can reduce the 24h uPro, decrease BUN and Scr in blood serum, and improve the renal function. As a result, the chronic renal failure progess is delayed. By depressing TGF-β1 protein expression in renal tissue, reducing the Smad2/3 protein expression, rising the Smad7 expression, and reducing TGF-β1RⅠ,RⅡmRNA expression, the ECM composition is reduced. Therefoe, the renal interstitial fibroblast is relieving by Shenshuaibaoshen Capsule, and chronic renal failure progress is delayed.
