AIM:To observe angiotensin-converting enzyme 2 (ACE2), transforming growth factors-β1 (TGF-β1) expression in renal tissues of adenine induced chronic renal failure rats, and the effects of Extract Ginkgo Biloba 761 (EGb761) on ACE2, TGF-β1 expression level.METHODS:Rats were randomly divided into EGb761 group, losartan group, integrated group and chronic renal failure control group (model group) after the chronic renal failure rat model was prepared. EGb761 group received EGb761 suspension (100mg·kg-1-·d-1), losartan group received losartan suspension (10mg·kg-1·d-1), integrated group received EGb761 (100mg·kg-1·d-1) and losartan (10mg·kg-1·d-1) suspension. And the normal control and model group received distilled water by gavage. The rats were sacrificed after 30 days'medication, blood, urine and kidneys samples were collected for kidney function, albuminuria and pathomorphology analysis. Immunohistochemistry was used to detect the amount of ACE2 protein. Real Time PCR was used to detect ACE2 mRNA and TGF-β1 mRNA expression.RESULTS:1. Model group serum creatinine (Scr), blood urea nitrogen(BUN),24-hour urine protein (24h MTP), kidney weight, kidney index were significantly increased than normal control group (P<0.01). Compared with the model group, Scr, BUN,24h MTP, kidney weight, kidney index of EGb761 group, losartan group and integrated group were significantly decreased (P<0.01).2. Immunohistochemistry showed that renal tissue ACE2 protein expression positive area and integrated optical density (IOD) of model group decreased compared with normal control group(P<0.01). Renal tissue ACE2 protein expression positive area and IOD of EGb761 group, losartan group and integrated group was significantly increased compared with model group (P<0.05).3. Real Time PCR results showed that the ACE2 mRNA expression of model group decreased significantly compared with normal control group (P<0.01); while the ACE2 mRNA expression of EGb761 group, losartan group and integrated group was significantly increased compared with model group (P<0.01).4. Real Time PCR results showed that the TGF-β1mRNA expression of model group increased significantly compared with normal control group (P<0.01); while the TGF-β1 mRNA expression of EGb761 group, losartan group and integrated group was significantly decreased compared with model group (P<0.01).CONCLUSION:1. Scr, BUN,24h MTP and morphology change of the kidney of the modeling rats indicate that the chronic renal failure models were successfully built.2. EGb761 protected the kidney function of adinine-induced renal failure rats maybe b increasing the ACE2 mRNA and protein expression and reducing the TGF-β1 mRNA expression3. Losartan can protect kidney function by many pathways, one is antagonizing AT1 whic is well proved now, maybe increasing the ACE2 mRNA and protein expression and reducing th TGF-β1 mRNA expression is another pathway.4. EGb761 and losartan could protect renal function separately, but these two drugs did nc have synergic effects when using together in treating chronic renal failure.
