Objective:To investigate the mechanism of Shenshuaifang on anti-renal interstitial fibrosis and retarding the progress of chronic renal failure by observing morphologic and functional changes of the kidney and detecting the protein contents of TGF-β1, AQP-2 and Bax in the kidney tissues of rats.Methods:Adenine was used to induced the renal interstitial fibrosis rat model. All animals were divided into 6 groups:normal group, model group, Shenshuaifang high dose group, Shenshuaifang low dose group, losartan group and combined drugs group(losartan and Shenshuaifang high dose group). Signs and general conditions of rats were observed.24-hour urine volume, urinary protein, serum Scr, and BUN changes of rats before and after the modeling and treatment were tested. Morphologic changes of kidney tissue were observed by hematoxylin and eosin staining of pathological section of the rats. Protein contents of TGF-β1, Bax and AQP-2 in renal tissue were detected with three-step method of immunohistochemistry.Results:Shenshuaifang can obviously improve the signs and symptoms of rats with renal interstitial fibrosis. Compared with the model group,24-hour urine volume, urine protein and protein contents of TGF-β1 and Bax in kidney tissue of treated groups were significantly reduced (P<0.01). Protein content of AQP-2 in treated groups were significantly increased compared with the model group (P<0.01).Conclusion:Shenshuaifang can reduce the 24-hour urinary protein and urine volume, decrease serum Scr, BUN, improve renal function, thereby delaying the progress of chronic renal failure. It can decrease interstitial fibrosis of kidney tissues by inhibiting TGF-β1 protein and probably have a relationship with reducing Bax protein that lessen the death of renal tubule cells, which intervene the renal interstitial fibrosis in certain extent. Also it can delay the progress of chronic renal failure by increasing AQP-2 protein levels in renal tissue.
