| Objective: To discuss the protective effect on dopaminergic neurons in substantia nigra of the Parkinson's disease model mouse treated by Electroacupuncture Scalp Point-through-Point therapy.Methods: The Parkinson's disease model mouse were modeled by stereotaxical microquantitative injection in the left corpus striatum with 6-hydroxydopamine and treated by Electroacupuncture Scalp Point-through-Point therapy.All the experimental mouse were divided into four groups: Normal Group,Sham-modeling Group,Model Group and Electroacupuncture Scalp Point-through-Point therapy Group: (1) Light microscope and transmission electron microscope detecting for the morphologic changes of substantia nigra and striatum positive neurons; (2) Immunohistochemical staining methods and RT-PCR for the morphologic,number changes of tyrosine hydroxylase positive neurons and the exppression of tyrosine hydroxylase mRNA; (3) In situ hybridization histochemistry was applied for testing expression of brain-derived neurotrophic factor mRNA;(4) RT-PCR for testing expression of dopamine transpotor mRNA; (5) RT-PCR for testing expression of Nestin mRNA.Results: (1) The total tyrosine hydroxylase positive neurons on the volume density,number density and light density of the substantia nigra of the destructive side in Model Group is obviously lower than that in Normal Group and Sham-modeling Group(P<0.05).Compared to Model Group,the relevant indexes of the Electroacupuncture Scalp Point-through-Point therapy Group after acunpuncture treatment are obviously enhanced (P<0.05);(2) Compared to Model Group, the expression of tyrosine hydroxylase mRNA is enhanced in Electroacupuncture Scalp Point-through-Point therapy Group(P<0.01);(3) The expression of brain-derived neurotrophic factor mRNA in the volume density,number density and light density in Model Group is obviously lower than that in Normal Group and Sham-modeling Group(P<0.05).Compared to Model Group,the expression of brain-derived neurotrophic factor mRNA is enhanced in Electroacupuncture Scalp Point-through-Point therapy Group(P<0.05);(4) The expression of dopamine transpotor mRNA in Model Group is obviously lower than that in Normal Group and Sham-modeling Group(P<0.01).Compared to Model Group,the expression of dopamine transpotor mRNA is enhanced in electroacupuncture Scalp Point-through-Point therapy Group(P<0.05);(5)The expression of Nestin mRNA in Model Group is obviously enhanced than that in Normal Group and Sham-modeling Group(P<0.05).Compared to Model Group,the expression of Nestin mRNA is enhanced in Electroacupuncture Scalp Point-through-Point therapy Group(P<0.05).Conclusions:The Electroacupuncture Scalp Point-through-Point therapy demonstrate the protective effect on dopaminergic neurons in substantia nigra of the Parkinson's disease model mouse. The protective mechanism is related to the effective of neuronal nourishment by brain-derived neurotrophic factor,acceleration of the synethis and metabolism of dopamine and the proliferation of nerve stem cell. |
