Protective Effects Of Banxia Xiexin Decoction On Gastric Mucosa Of The Rats With Hp Infection And Its ERK Signaling Transduction Mechanism

Objective:By observing the reparation effects of Banxia xiexin Decoction on treatment of model rats with Hp infected gastric mucosal lesion, the contents of EGF and PCNA in gastric mucosa of rats and the expression of key factors(EGFR, P-ERK1/2, CyclinD1)in ERK signal transduction pathway, this thesis aims at exploring the molecular biology basis of banxia xiexin decoction on the impairment of gastric mucosal lesion and confirming that banxia xiexin decoction encourages the reparation of the scathing gastric mucosa in Hp infected rats by cell proliferation based on the theoretical bases of "Strengthening the body resistance" in TCM, thus plays an effective role in treatment of Hp related gastritis.Methods:73rats (SPF, C57BL/6) were randomly allocated into7groups, namely model, quanfang ganwen, kuhan, xinwen, positive control and blank group. The animal model of rats with Hp infected gastric mucosal lesion were prepared by gavage of Hp bacterium solution. The banxia xiexin decoction and the disassembled prescriptions were used as the test factors. The result was compared with that of the standard triple therapy. The histomorphologic changes were observed under light microscope. The contents of EGF and PCNA in tissue of gastric mucosa were detected by enzymelinked immunosorbent assay. The expressions of EGFR, p-ERK1/2and CyclinD1were detected by method of Real-time PCR and Western blot.Results:(1)After modeled, the inflammatory cell infiltration and the extent of glands atrophy of the gastric mucosa in the model group has been improved. The difference comparing with the blank group (p<0.01) is significant. When comparing the inflammatory cell infiltration, the extent of glands atrophy of the gastric mucosa in rats of the quanfang group, kuhan group and positive control group with that of the model group and xinwen group, the significant difference exits(p<0.01). In ganwen group, the inflammatory cell infiltration and the extent of glands atrophy of the gastric mucosa in rats has also been improved (p<0.05).(2)The content of EGF in scathing gastric mucosa of the Hp infected rats was increased in quanfang group, positive control group, ganwen group, which was obviously higher than that in the model group, xinwen group and kuhan group(p<0.05or p<0.01), however, the comparison of the difference of ganwen group and quanfang group has statistically significance (p<0.05).(3) The contents of PCNA in scathing gastric mucosa of the Hp infected rats was increased in quanfang group, positive control group, ganwen group and kuhan group, which was obviously higher than that in the model group and xinwen group(p<0.05or p<0.01). However, when comparing the ganwen group and kuhan group with quanfang group, the difference has statistical significance (p<0.05).(4)The expression of EGFR, p-ERK1/2and CyclinDl in scathing gastric mucosa of the Hp infected rats was up-regulated in quanfang group, positive control group and ganwen group, which was significantly different with that in model group (p<0.01). The difference between kuhan group, ganwen group&xinwen group and quanfang group has statistical significance (p<0.01or p<0.05).(5) Quanfang group of banxia xiexin decoction has an equal effect compared with positive control group in improving the inflammatory cell infiltration and the extent of glands atrophy of the scathing gastric mucosa of the Hp infected rats, the total score, the extent of EGF&PCNA in the scathing gastric mucosa of the rats, the expression of key factors of EGFR, p-ERK1/2and CyclinDl in upregulating ERK signaling transduction pathway. Conclusion: (1)Banxia xiexin decoction can significantly decrease the degree of the inflammatory cell infiltration and the extent of glands atrophy of the scathing gastric mucosa of the Hp infected rats, thus improving the situation of the histomorphological damage in gastric mucosa.(2)Banxia xiexin decoction can improve the extent of EGF and PCNA in scathing gastric mucosa of the rats, promoting cell proliferation and repair of the gastric mucosal.(3)Banxia xiexin decoction can promote the ERK signaling transduction by up-regulating the expression of EGFR in scathing gastric mucosa of the rats, thus achieving the aim of promoting the cell proliferation and repair of the gastric mucosa. The specific segments are as follows:the intervention of banxia xiexin decoctionâ†'the synthesis of EGF in Gastric Mucosaâ†'the expression of EGFR in cell of gastric mucosa, key enzyme of ERK phosphorylation, control of CyclinD1â†'cell proliferation of gastric mucosa.(4)The effect of banxi axiexin decoction is superior to the disassembled prescri-ptions in repairing the scathing gastric mucosa of the Hp infected rats, which manifested the "comprehensive effect" of comprehensive treatment. The ganwen group has obvious effect on cell proliferation and repair of the gastric mucosa, inferring that ganwen group is the major components of banxia xiexin decoction for its effect of "Strengthening the body resistance".(5)Banxia xiexin decoction has an equal effect compared with the standard triple therapy in repairing the scathing gastric mucosa of the Hp infected rats.
