Banxia Xiexin Decoction recorded in "Treatise on Febrile Diseases", with antiemetic Jiangni and Xiaopi Sanjie effect. From the perspective of modern medicine, the formation of epigastric fullness syndrome is related to disorder of gastrointestinal motility. And the weakening of gastrointestinal motility has an especially strong correlation. Gastrointestinal motility disorder of gastrointestinal disease is many important clinical manifestations and pathogenesis. In recent years, the researchers have done a lot of research about Banxia Xiexin Decoction and its regulating effect on gastrointestinal movement. It has significant effects on regulating gastric electric rhythm, enhancing gastric motor tension, promoting gastric emptying and intestinal propulsion.Based on the knowledge we just mentioned above, we carried out the study on Banxia Xiexin Decoction and its disassembled prescriptions regulating the mechanism of gap junction of gastric motion. The paper conclude two parts:In the first part, we reviewed the literatures of the pharmacological study and clinical application of Banxia Xiexin Decoction’s characteristics and its disassembled prescription drugs, and research progress of gastrointestinal ICC gap link communication and link protein Cx43, protein kinase C.To provide a theoretical basis for clinical application, Banxia Xiexin Decoction and its disassembled prescription drug pharmacology research is deepening day by day, and a great deal of achievements has been gained about the mechanism of the regulation of gastrointestinal motility and the compatibility mechanism etc. Banxia Xiexin Decoction widely is used in clinic to treat many diseases, such as acute and chronic gastroenteritis, peptic ulcers, gastro esophageal reflux disease, functional dyspepsia, etc. In semiology, epigastric fullness is very similar to Gastric Dysrhythmia Syndrome (GDS). The major symptoms of GDS include nausea, vomiting, abdominal distension, loss of appetite, etc. All the symptoms mentioned above all familiar in many gastric functional diseases and nerve regulation disorder. In clinic, GDS can be divided into three kinds, the tachygastria, bradygastria and gastric dysrhythmias. SMC changed in network structure and function has pathophysiological significance of GDS. In the gastrointestinal tract, GJ is a special channel between the SMC and ICC cell mediated electrochemical information exchange.Cx43 is the most significant link of connexin gap, and the current study includs its structure and function, in the gastrointestinal distribution and mechanism etc. As an important intracellular signal transduction system, protein kinase C plays an important role in the transmembrane information transmission. The study of Banxia Xiexin Decoction and its effect on the expression of Cx43 and PKC-α, is of significance to research the mechanisin of gastrointestinal motility disorder in the further.In the second part, we combined modern mathematical statistic methods and recipe evolvement rule, divided Banxia Xiexin Decoction into a Xinkai group, a Kujiang group, a Ganbu group and any three groups in two groups of binding. Rat models of electrogastric dysrhythmias were established, and were used to evaluate the index of Gastric electrophysiology before randomization. Each treatment group lasted administration for 4 weeks by the same drug concentration of unequal volume administration. Each group included 10 rats and was analyzed for gastric electrical parameters after administration. And the expression of Cx43、PKC-α and their mRNA in the gastric tissue of rat in each group was analyzed by immunohistochemistry and real-time fluorescent quantitative PCR. According to the experimental results, it was analyzed in the efficacy and compatibility characteristics of side group. And the result revealed the bidirectional adjusting gastric motion mechanism of Banxia Xiexin Decoction. The results of this study are as follows:Banxia Xiexin Decoction and all the components groups have effects on correcting the disorder of gastric electric rhythm in different degrees. Compared with the model group, all the components groups had observably reduced the coefficient of variability of the model group’s slow wave eletrogastric rhythm. The result suggested that all the components have effects on adjusting gastric electric rhythm. And the Xinkai and Kujiang group had the lowest coefficient of variability of the model group’s slow wave eletrogastric rhythm. As was it, prompt symplectic herbs with bitter medicine compatibility could better adjust gastric electric rhythm abnormalities.Semi quantitative analysis showed that each drug group had decreased the expression of Cx43 to some extent. The group containing herbs in pungent and sweet flavor had markedly decreased the expression of Cx43. The result indicated that each drug group had decreased the expression and distribution of Cx43 in different degrees compared with other groups. The results of factorial analysis showed that there were interaction and restriction factors among each drug groups. It suggests that there is the synergistic effect between the drug groups.The expressions of Cx43 and Cx43mRNA in the model group were significantly increased compared with other groups. Banxia Xiexin Decoction and its components groups had decreased the expression of Cx43 and Cx43mRNA to some extent. Among which the group containing herbs with pungent and pleasant flavor had best effects on the index. There was a statistically significant difference between the normal group and the model group.The expressions of Cx43 in the model group were significantly increased compare with other groups. But the expressions of PKC showed a reverse trend. Banxia Xiexin Decoction and its components groups had decreased the expressions of Cx43 and increased the expressions of PKC-α and PKC-αmRNA to some extent. Therefore it was used to adjust gastric electrical rhythm group of disorders dysrhythmias and improve gastric motility function.To sum up, the group containing herbs in sweet flavor is the major component in regulating gastric electric rhythm disorder. The group containing herbs in pungent and sweet flavor is the major Jingfangfangyuan in suppressing the content of Cx43, and the xinkaikujiang group is the major Jingfangfangyuan in promoting the content of PKC-α. The mechanism of Banxia xiexin Decoction adjusting gastric motility maybe relate to its function of decreasing the expression of Cx43 and Cx43mRNA and increasing the expressions of PKC-α and PKC-αmRNA, as to adjust gastric electrical rhythm group of disorders dysrhythmias and improve gastric motility function.
