Research of traditional Chinese medicine (TCM) can not explain Chinese medicine holistic connotation well with using the "dialectical theory of governance" method. As the rapid development of bioinformatics technology, under the background of big data, network pharmacology and system pharmacology arises at the historic moment. These modern technologies can not only forecast the mechanism and active components in TCM, but also explain the new indications of TCM, which transforms disadvantages into the advantages, to promote the modernization of TCM. As a model organism, Drosophila plays an important role in many research areas. Without lymphocytes that mammalian have, gut and innate immunity construct the important barrier to resist foreign invasion in Drosophila. To explore the therpeutic function of TCM in intestinal inflammation and chronic inflammation, we studied the influence of herbs on Drosophila intestinal immune function when feeding metal ions, proinflammatory cytokine, oxidizing agent and high salt; we also studied the Drosophila innate immune effects of TCM on injection of Beauveria bassiana (B. bassiana), Serratia marcescem (S. marcescem) and Staphylococcus aureus (S. aureus). As the direct inducing factors of metabolic syndrome and type 2 diabetes (T2D), insulin resistance (IR) severely impairs the human health. To explore a unique efficacy single TCM which can prevent and cure insulin resistance, we constructs the insulin resistance model induced by high sugar diet use Drosophila, and via this model to select TCM with mitigating effect on insulin resistance. The main results obtained are as follows:1. The number of TCM increased the Drosophila survival rate after feeding metal ions (CuSO4, ZnSO4 and KA1(SO4)2-12H2O) were 16,56 and 27, respectively. After ingestion sodium dodecyl sulfate (SDS) and paraquat (PQ),15 and 21 kinds of TCM enhanced the survival rate.12 kinds of TCM improved the survival rate after ingesting NaCl for 6 days.2. Asparagus cochinchinensis (AC) extract could increase the survival rate of feeding metal ions in Drosophila, reduce the amount of gut epithelial cell death, and alleviates gut cell morphology. These results indicate that AC extract has a protective effect on metal ions-induced gut injury in Drosophila.3.6 kinds of natural products (NPs) have better drug likeness compared with anti-inflammatory agents. We speculated that UNPD133185, UNPD43533, UNPD77220, UNPD135865, UNPD68648, UNPD96599 these 6 kinds of NPs which collected from Universal Natural Products Database are the potential active constituents that mediate gut injury.4. Oldenlandia diffusa (O. diffusa), Polygonatum odoratum (P. odoratum) and Polygonatum sibiricum (P. sibiricum) significantly increased the survival rate after injecting B.bassiana.5. In this study, we successfully constructed the insulin resistance model induced by high sugar diet. The model has the characteristics of good reproducibility, simple modeling, low cost and so on.6. The extract of Ampelopsis japonica (AJ) has a significant alleviation of Drosophilia growth and lipid accumulation on high glucose diet induced insulin resistance; at the same time, 7 kinds of chemical constituents in AJ can interact with lipid binding signal pathway. This suggested that AJ can achieve the aim of remitting the insulin resistance through the regulation of lipid accumulation.
