The second-generation Brush-Type CSP (DNB-Leucine CSP) had been researched in this thesis, including the synthesis of DNB-Leucine CSP and the column packing for analytical and preparative scales. The efficiency of these columns also had been tested, The results and the mechanism had been discussed basis on the experimental results.Through the experiment, we found the best conditions for packing column. The condition for analytical column was obtained by maintaining packing pressure 35 Mpa for two hours, and the condition for preparative column was obtained by maintaining packing pressure 20 Mpa for two hours.The theoretical plate of analytical column is related with the flow rate of mobile phase, the type of sample and the injection quantity. The best the oretical plate was obtained while the injection quantity of toluene was restricted in 20μg,with the flow rate of 1ml/min, hexane as the mobile phase. For the preparative column, the best theoretical plate was obtained while the injection quantity of toluene was restricted in 100μg, with the flow rate of 10ml/min, hexane as the mobile phase. The method of testing the efficiency of DNB-Leucine CSP was proposed first time, and the efficiency could be explained with Van Deemter equation.Both the analytical and preparative DNB-Leucine CSP columns, the two enantiomers, clenbuterol and N-(1-(1,2dihydroacenaphthylen-5-yl)pent-4-enyl)-3,5 -dinitrobenzamide had been separated. We also calculated the load ability of prepa -rative DNB-Leucine CSP column.
