Purpose:To observe the Yi Jing Fang method of adenine-induced infertil-ity deficiency rat sperm density and motility, serum hormone levels and theimpact of claudin/occlaudin protein expression.Materials and Methods:48healthy SPF Wistar male rats,3months old,weighing180±20g,were randomly divided into control group, model group,Xuanju group,Yi Jing Fang groups of12.In addition to the control group,theother groups are adenine1mL/(100g· d) dose gavage12days;13days fromthe beginning of the control group and model group were fed with normal sa-line, Xuanju group to Xuanju capsules aqueous gavage, Yi Jing Yi Jing FangFang group to soups gavage once daily for20days. After the end of the ex-periment with10%chloral hydrate anesthetized rats,blood10mL under steri-le conditions in the abdominal aorta,the removal of the testis and epididy-mis,All rats were killed after the end of the experiment,the removal of thetesticles, sperm analysis system using computer count sperm density and mo-tility;using enzyme-linked immunosorbent competition law and doubleantibodysandwich enzyme-linked immunosorbent serum testosterone were detected (T),estradiol alcohol (E2) and serum inhibin B(INHB) level,using immunohistoche-mical assay claudin/occlaudin protein structure was observed by HE stainingof testicular tissue under a microscope.Results:1. compared with the control group, model group sperm motility,sperm dens-ity,T and INH B levels were reduced, elevated E2levels,reducing claudin/occlaudin protein expression,the difference was statistically significant(P<0.01);2. Compared with model group,Xuanju group and Yi Jing Fang group sperm mot-i lity,sperm density,T and INH B levels were elevated,reducing E2levels inc-reased claudin/occlaudin protein expression,the difference was statistical-ly significant (P<0.05);3. Compared with Xuanju group,Yi Jing Fang group sperm motility,sperm dens-ity, T, and slightly elevated levels of INH B, E2levels slightly lower,the difference was not statistically significant(P>0.05);claudin/occlaudinprotein increased, the difference was statistically significant (P<0.05).Conclusion:1. Adenine can reduce sperm concentration,sperm motility and serum hormone T,INHB levels,elevated E2levels;while the expression of claudin/occlaudinprotein is reduced;2. Yi Jing Fang can improve sperm density deficiency of adenine-induced ratmodel,sperm motility and serum hormone T,INHB levels,lower levels of E2;whi-le the expression of claudin/occlaudin protein increased.
