Effect Of Integrated Traditional Chinese And Western Medicine On Methylation Of MMP - 9 Gene Promoter In Patients With Phlegm Blood Stasis Syndrome

Objective:To study the relationship between the gene promoter methylation level of matrix metalloproteinase-9(MMP-9) and ACS, and observe the clinical efficacy of combined treatment of traditional chinese and western medicine in treating ACS(phlegm and blood stasis type).Methods: Selected 60 ACS patients(30 unstable angina pectoris patients and 30 miocardial infarction patients) dialectical as phlegm and blood stasis type as ACS group, 40 normal people were selected as the normal group.All of the patients in ACS group after PCI treated with Traditional Chinese Medicine.Treatment for 1 months.MMP-9 promoter DNA methylation was detected in ACS patients before and after the treatment and normal group by methylation-specific PCR, Comparing the demethylation level between the normal group and ACS group,the treatment group and the ACS group.Results:1.AS compared with control group,there was the lower level of the average density of the DNA methylation of the MMP-9 genepromoter in the ACS patients(26.7%VS45%,P<0.05).2.There was the significantly higher level of the average density of the DNA methylation of the MMP-9 genepromoter in the treatment group after treatment of PCI and traditional Chinese Medicine(36.7%vs26.7%,P<0.05).Conclusion:The incidence of ACS associated with its low methylation status of MMP-9 gene promoter.Combination of Chinese traditional and Western Medicine has obvious therapeutic effect on Phlegm and blood stasis type ACS, its regulating effect on the gene promoter methylation level may be one of the mechanism of effect.
