Effects Of Shenqi Xiezhuo Decoction On Expression Of MMP-1 And TIMP-1 In Renal Tissue Of Adrenaline Interstitial Fibrosis Rats

Purpose:To research Shen Qi Xiezhuo drink on adenine-induced chronic renal failure in rats kidney tissue matrix metalloproteinase 1(MMP-1), matrix metalloproteinase inhibitor 1(TIMP-1) gene expression.Material and method:SPF male SD rats were 56.Divided the rats into normal group and model group by balloting.There are 10 normal rats,and the rest are in model group. The model of chronic renal failure in rat was set up by weight in Yokozawa method.Each rat in model group was given adenine daily at a concentration of 2.5%,200 mg/kg,similarly to the volume of normal water as a control.Verificated model after experiments were performed 4 weeks.Hence,the remaining rats were randomly divided into model group,shenqi Xiezhuo drink high-dose group,the conventional-dose group,losartan group and model group level based on their weight.The rats were fed once a day,which high-dose group was given shenqi Xiezhuo decoction of 20ml/kg,conventional dose group were given shenqi Xiezhuo decoction of 10ml/kg,losartan group were given losartan of 9mg/kg,model group and the control group of rats were fed pure water of 10ml/kg.8 weeks later,anaesthetized the all rats,and collected abdominal aortic blood immediately,then detected Scr、BUN、ALT in ELISA and Detected renal tissue MMP-1, TIMP-1 gene expression using quantitative PCR.Results:Scr、BUN、ALT in serum of all rats has been declined after being treated by Shen Qi Xiezhuo decoction,compared with before.(1)Scr, BUN in High-dose group were significantly lower than other groups(p<0.05),while conventional-dose group had no significant difference with the losartan group,and lower than the model group(p>0.05).(2)MMP-1 m RNA expression in high-dose group was significantly higher than conventional-dose group,losartan group and model group(p<0.05);while conventional-dose group had no significant differencewith the losartan group(p>0.05),but higher than the model group(p<0.05).TIMP-1 m RNA expression in high-dose group was significantly lower than others(p<0.05),Conventional-dose group had no significant difference with the losartan group(p>0.05),but lower than the model group(p<0.05).Conclusion:It was useful to treat renal failure in rats by drinking Shen Qi Xiezhuo decoction.It could improve their general symptoms,and delay the development of the disease.In this studies, we can see,the decoction can slow progression of CRF and protect the kidneys by promoting MMP-1m RNA expression and inhibiting TIMP-1m RNA expression.
