Circulation Line Water Prescription Anti-rat Renal Interstitial Fibrosis In Rats

OBJECTIVE:To study the effects of the Huayu Xingshui recipe on preventing the renal interstitial fibrosis in rats.METHODS:The rats were randomly divided into unilateral ureteral obstruction(UUO) group and sham operation group.The former group were furtherly randomized into model group,Huayu Xingshui recipe group (Chinese herbal group)and Captopril group.After administration of placebo, Huayu Xingshui recipe and Captopril respectively for 7days,14 days,21 days and 28 days,killing the rats and taking out the obstructed kidney,the collagen,MMP-9,TIMP-1,CollagenⅢmRNA of the renal interstitium were examined by semi-quantified image analysis system.RESULTS:Compared with the sham operation group,The collagen,the expression of MMP-9.TIMP-1 and CollagenⅢmRNA in renal interstitium of UUO group were increased significantly(P＜0.05orP＜0.01).In comparison with the model group,After 7days treatment,collagen in the renal interstitium was decreased in Huayu Xingshui recipe and Captopril group. The expression of TIMP-1 and CollagenⅢmRNA were decreased(P＜0.05), too.But,the expression of MMP-9 had no significant differrence between the model group and Chinese herbal group or Captopril treatment group.After 14days,21days and 28 days treatment,Huayu Xingshui recipe group and Captopril group decreased collagen,the expression of TIMP-1 and Collagen ⅢmRNA(P＜0.01),but not to the level of that in sham operation group(P＜0.01).increased the expression of MMP-9 in renal interstitium (P＜0.05).CONCLUSION:Huayu Xingshui recipe treatment can decrease collagen,TIMP-land CollagenⅢmRNA,and increase MMP-9 in renal interstitium.Huayu Xingshui recipe has certain effecct on preventing the renal interstitial fibrosis.
