Xuejie Sanqi Hydrogel Patch whose Prescription was from hospital preparations, was comprised of Draconis Sanguis, Notoginseng Radix et Rhizoma, Olibanum, Myrrha, Carthami Flos, Dipsaci Radix, Cibotii Rhizoma, Acanthopanacis Cortex, Pyritum, Cervi Cornu Pantotrichum and Borneolum Syntheticum,11 herbs. With blood circulation, reduce swelling and analgesic effect, so it’s mainly used for the treatment of bruises, broken ribs fractures, bruising swelling, and could achieve better clinical curative effect. This research studied the formulation selection, pieces quality standards, the extraction process, concentrate and dry grinding process, preparation molding process, pilot production, quality standards, stability etc, guided by the theories of TCM, combined with analysis of prescription medicine, the use of modern pharmaceutical methods and technology.Meeting the demands of the chemical, clinic and physical properties of the drugs, and the experimental results, the form of formula finally was decided as cataplasm. Under the relevant requirements of medicinal herbs and decoction pieces according to the 2010 edition of Chinese pharmacopoeia,11 herbs in the prescription had carried on the appraisal and pretreatment, in order to ensure its source stability and quality control. Through literature research, pharmacodynamics screening and preparation feasibility study, the process route was determined. That was Notoginseng Radix et Rhizoma, Olibanum, Myrrha, Carthami Flos, Dipsaci Radix, Cibotii Rhizoma and Acanthopanacis Cortex with water extraction, Draconis Sanguis, Pyritum and Cervi Cornu Pantotrichum with powder medicine (extremely fine crushed separately), Borneolum Syntheticum with alcohol soluble medicine. Through orthogonal experiment, the best water extraction process parameters was established, with hydroxysafflor yellow A, notoginsenoside R1, ginsenoside Rgl, asperosaponin VI, ginsenoside Rbl and ratio of extraction as index components.In the study of preparation for molding process, the appearance description, initial bonding strength, endurance bonding strength, and peel strength were taken as indexes. Based on the result of a single factor experiment, the formula for the Sanqi Hydrogel Patch was optimized by central composite design-response surface methodology. The optimal ratio of the prescription was as follows:ViscomateTM NP700-Carbomer 940-PVPK90-kaoline-dihydroxyaluminum aminoacetate (1.86 ：1.48: 0.49:0.5:0.16). And transdermal properties, preservatives, drug loading, were investigated to determine the formulation process.In the quality specification study, TLC identification methods of Draconis Sanguis, Notoginseng Radix et Rhizoma, Dipsaci Radix, Cibotii Rhizoma, Cervi Cornu Pantotrichum, Borneolum Syntheticum were set up. And the rules of preparation had been checked, content determination methods of hydroxysafflor yellow A, notoginsenoside R1, ginsenoside Rg1, asperosaponin VI, ginsenoside Rb1 and dracorhodin were established.
