In order to breed new varieties of high quality of Auricularia polytricha, this study used single spore cross breeding.The parentstrain is yellow strains of AP10 and AU2 ,and white strains of APW822. Three parents have distantly related and complementary agronomic. Select robust and dense mycelial growth single spore which have different mating type factor that can pair to form clamp connection from all single spore strains of parentstrain to pair. The main results are as follows:1. Screening parent strainsThe screening strains based on strains preserved by Sichuan Academy of Agricultural, By ITS sequence analysis of 13 strains, the phylogenetic tree was established. With statistical analysis of DNA fingerprinting, the clustering based on ISSR similarity map was established, and finally selected distantly related, complementary agronomic traits of three strains as parents strains, that AP10, AU2 and APW822.2. Research about the polarity of Auricularia polytrichaMost current research shows that the polarity is the bifactor hetenthallism, but some early scholars believe that the polarity is monofactor hetenthallism, for this phenomenon, this study used three matching method to study the polarity of single spore of three parental strains, and isolated mononuclear protoplasts of three parental strains, each parent of two mating types of mononuclear protoplast was separated and its mating type was measured. Suitability test of the above results, test results show that the actual strain of each mating type consistent with the theoretical number, may be considered the mating type of Auricularia polytricha is bifactor hetenthallism.3.Genetic analysis of hybridsThrough the experiment on hybrid antagonists, determine mycelial growth rate, the ears experiment and the establishment of clustering patterns based on ISSR similarity coefficient, Some conclusions was found : (1) The yield of 11 hybrids based on AP10 and AU2 matching are more than the yield of parental strains and the yield of 1 hybrids based on AP10 and APW822 matching is more than the yield of parental strains .The hybrids based on AP10 and AU2 matching are superior to hybrids based on AP10 and APW822 matching; (2) Hybrid AU55 have Several advantages,such as mycelium dense, high yield, strong disease resistance, morphological characteristics between the two parents and distantly related with other hybrids, AU55 is a better quality strains; (3) Between mycelial growth rate and intensity and the production of strains are positively correlated, but not absolute; (4) Genetic stability of organization separation species of hybrids is superior to the multispore isolation of species.
