Ap10and ApHP are major cultivars in China and the yield of the two varietieshad declined in recent years, which brought huge loss to peasants. We can getrejuvenation strains by a series of methods, which fill in a gap in developing newcultivars and provide good parent for breeding. The major studying results are asfollows:We got366rejuvenation strains through three methods: protoplast regeneration,tip isolation and tissue isolation.17strains had increased yield and39strains hadstrong antiviral ability. Through comprehensive analysis of dry matter yield,agronomic traits and disease resistance, we screened8excellent rejuvenation strains.During regeneration of Auricularia polytricha protoplast, the growing rate ofheterokaryotic isolations was very high in early period. Yield increase rate ofApHPP32and Ap10P78were27.48%and13.09%respectively. They were the bestrejuvenation strains with good agronomic traits and disease resistance. It wasconcluded that the protoplast regeneration rejuvenation effect of ApHP was betterthan Ap10. Degenerative strain Ap43was hard to fruit, which verified that ApHP andAp10were not degenerative strains.ApHPt4and Ap10t8were isolated fom tip of the hyphae, whose yield increaserate were13.93%and14.14%seperately. The two rejuvenation strains, ApHPt4andAp10t8, were from twice tip isolation with the treatment of9h under45℃and40℃respectively. The effect of high-temperature stimulating on Ap10was more significantthan that on ApHP.The yield increase rate of the best rejuvenation strain, Apf4f6, which had goodagronomic traits, was13.83%and infection rate was reduced to13.33%. Apf1f9andApf1f6were were excellent resistant parents for breeding because of the low infectionrate with3.33%and0respectively. Ap1082f and Ap1081f4were good resistantstrains isolated from tissue of Ap10. It was concluded that the effect of tissue isolatingon ApHP was better than that on Ap10. The dry matter yield of rejuvenation strains gotten from three methods waspositively correlated with CMC enzyme activity and xylanase activity, correlationcoefficient was0.605and0.625respectively. The genetic materials of these strainswas consistent with original one and virus hadn't found yet. Compared by yieldincrease rate, the protoplast regeneration method was the best one for rejuvenation.
