Ganoderma lucidum is a treasure in our country's medicine. it has been called'fairy herb'or'lucky herb'for a long time. But the Spore of Ganoderma lucidum is difficult to germinate in the artificial condition. It is unable to obtain mononuclear strain for Ganoderma lucidum breeding,which make conventional methods of breeding develop slowly. In this study the hybrids of Ganoderma lucidum is obtained by protoplasting monokaryotization technology, The result show that some hybrids have significant hybridizating advantages. Several conclusions were made as follow:First,In this study, biostatistics and molecular biology is used to select the parents who have distant genetic distance and complementary agronomic traits, Finally five strains were selected as the parents in hybridition test .Secondly, In the process of the protoplast regeneration,it was found that the germination of different mating type is not same. The time of different mating type has obvious sequencing,One of mating type concentrated in the early stage ,And another concentrated in the late . The strains of different mating type also have differences on the mycelial morphology.In this study, 81 protoplast regenerating mononuclear strains were obtained through monokaryotization by protoplasting technique, Among them, seven mononuclear strains of different mating type was determined. On the base of hybrid breeding was carried out, The results showed that 24 hybrids which is different from parents were obtained.Hybrids were verified using a variety of tested , which determined that hybrids is different from parents.the tested were as following: a preliminary study on the impact of temperature and pH value on the hybrids mycelial growth, And did a series of experiments about antagonist, and mycelial growth rate of hybrid and the parents in cultivating materials. It is found that the hybrid named 30+2-7×24-7 and 06-6×30+3-7 are the fastest mycelial growth rate at 0.9380 cm/d and0.460 cm/d respectuely.The results showed that the hybrids have significant differences with parents.Molecular biology metheds ware used to identify hybrids,It found that the genetic material of hybrids is from the two parents.The polysaccharide content of fermentation,mycelium and fruitbody and production were tested .The data show that the hybrid named 30+2-7×24-7 has significant hybridizating advantages on polysaccharides contect and output with the parentsNo correlation between the production and mycelial growth rate of hybrid.and also no positive correlation was found between the traits of hybrids and parental genetic distance .
