In this study, Ganoderma lucidum and Ganoderma sinense were chosen for monokaryotization by protoplasting technique. Then, mono-mono mating tests between protoplasted monokaryons of Ganoderma lucidum and Ganoderma sinense were carried out. Eight hybrids were obtained and identified by morphology, molecular markers, analyzing biological characters, and medical compounds, and some of them had significant hybridizating advantages which convergence the excellent qualities of the two parents.1. Selection of parents and formation of protoplasted monokaryonsSeven parent heterokaryon strains of Ganoderma lucidum and Ganoderma sinense were chosen for monokaryotization by protoplasting technique in different concentration of lywallzyme and treat time. Two types of monokaryons were obtained in each parent strains.2. Mono-mono mating tests and hybrids obtainedDifferent mating genotypes of protoplasted monokaryon strains were confirmed,and there were858paired combinations. Eight hybrids were obtained through mono-mono mating tests between monokaryons of Ganoderma lucidum and Ganoderma sinense, the matching rate was0.9324percent.3. Identification of hybridsHybrids were initially identified based on somatic incompatibility tests and microscopes of clamp connections and stained nuclea. Furthermore, ITS-RFLP, SRAP and RAPD were carried out, and the results showed that hybrids were different from the parent strains.4. Research on biological charactersTemperature, pH, growth rate on PDA and cultural media were studied in the test. The results showed that hybrid03+1-51x07-5had the fastest growth at30℃on PDA medium accounting for4.68mm/d; the quickest growth rate of03+1-18×07-42was4.50mm/d at pH7;12-71×05-1had greatest rapidity10.05mm/d in growth tubes, and12-77x05-1had greatest rapidity3.36mm/d in cultural media respectively. The results also indicated that there were significant differences between parent strains and hybrids in these ways.5. Determination the polysaccharides and triterpenoidHybrids and parent strains were evaluated by extraction and determination of the polysaccharides and triterpenoid. The analysis result showed that the total polysaccharides and triterpenoid of hybid12-77x05-1was significantly superior to parent strains, and higher than other hybrids at the both.
