This study using single spore hybridization technique obtained31Ganoderma lucidum hybrid strains successfully by comparative analysis of24parents stains comparatively in genetic relationship, effective component content and agronomic traits. To complete the comprehensive evaluation of hybrid strains obtained and screened3new superior varieties with obvious heterosis.1、The genetic diversity of24parents strains using ISSR molecular marker technique were analyzed, the results show that:according to the similar coefficient matrix and dendrogram of cluster analysis based on ISSR markers when the similarity coefficient is0.642,24strains is divided into3groups, No.14and15strains as a group (group Ⅱ); No.18、20and22as a group (group Ⅲ); the rest of the strains clustered into one group (denoted as I)2、The main effective component analysis results of24parents strains showed that G.10001、G.10005、 G.s0018, and G.10025is the polysaccharide and triterpene content higher varieties, the polysaccharide content of G.10001was the highest, reaching3.3871%, triterpene content as high as1.5432%; two content of strain G.10003were low, considered weak strain.3、According to the principle of distant genetic distance and complementary agronomic traits, finally G.10001, G.10003, G.10005, Gs0018, G.10025were selected as parents stains.4、The mating type of5Ganoderma Lucidum parents strains were identified by the mycelia performance of two monokaryon strains in the spore-crossing test and the presence of true or false clamp connection。 Based on the large number of monokaryon strains and accurate identification of the mating type,2strains having special mating type (ArB1and ArB2) composed of A factor new and the parental type B were obtained, and according to the round of the mating reaction, the five tested strains of mating genotype were inferred, respectively A1A2B1B2、A2A3B1B2、A1A2(A4) B2B3N A1A3B2B4and A5A6(A7) B5B6。5、31hybrids were obtained successfully by single spore hybridization technique,and the antagonistic reaction validated that these hybrid strains were different from their parents preliminarily.6、The cultivation of Hybrid strains test out4stains can not form a fruiting body, further comprehensive evaluation to the rest of the27hybrid strains showed that:1-6×14-32,1-4×14-5,11-11×14-7is a new variety with obvious heterosis.
