Using the modern extraction, separation active ingredients from sporoderm-broken ganoderma spores. With papain and activated carbon on Ganoderma lucidum polysaccharides in protein, pigments were isolated and used Ganoderma lucidum polysaccharides on the determination of antioxidant capacity. The main contents and conclusions are listed as follows:1. The extraction method of polysaccharide from Ganoderma Lucidum were systematacially studied, the method of leaching with hot water, ultrasonic-assisted extraction, supercritical CO2fluid extraction and enzymatic extraction were investigated respectively, and these processes were optimized, the advantage and shortcoming of these technology were compared from the extraction yield, and economic benefit. The results showed that:the extraction yield of lentinan extracted by hot water was 1.79%,, extraction temperature was 95℃, the Lentinus edodes mass to water was 1:25, extraction time 2h. the extraction yield by enzymatic extraction was 2.38%, mass ratio of proteinase to Ganoderma Lucidum was 1.5:100, the material was treated in solution about pH4.6,at 45℃for 45min, then extracted at 95℃for 2h. The extraction yield supercritical CO2 fluid was 1.81%, the optimal extract conditions were as follows:temperature of extraction was 50℃. pressure of extraction was 15mPa,the quantity of cosolvent 0.4ml/g for 4.5h.The extraction yield by ultrasonic-assisted extraction was 2.18%, tultrasonic time 90min, extraction temperature was 80℃, power 1200W。2. Using the activated carbon to decolorize the crude polysaccharide from Ganoderma Lucidum.The optimal decolorization duration, temperature and decolorant quantity were50℃, 1h and 1%, pH3.respectively,Under these conditions, the decolorization rate was over 85.1% and 86.2% of polysaccharide was recovered.Papain was used to remove protein, and the optimum conditions including 0.8:1 papain, temperature 50℃, pH5.5,60min. This method may increase the removal rate of protein to 89.18%。3.Through four different systems (hydroxyl radical·OH, superoxde anion O2-·, DPPH·, reducing power) of the broken spores of Ganoderma lucidum polysaccharides on the antioxidant properties. The results show that, it has strong antioxidant activity.
