Breast cancer is one of the most common carcinoma of women. The over view to breast cancer has been changed:at present,we think breast cancer is the disease all over the body at first,the mass of breast is the concentration of the disease. So the treatment to breast cancer is first focus on the systemic treat. Although we have used chemotherapy,radiotherapy and endocrinotherapy,there is no obviously improved in survival. We should look for more effective treatment. In the past experiment,we had found there is more CD40 on the breast cancer cell line MDA-MB-231 and navel vein endothelia cells. So we design this experiment to study the effect of CD40-ligand to breast cancer.The first part:M231 and ECV cells are incubated and examined CD40 using flow cytometer. M231 and ECV cells are incubated with ADM and CD40-ligand. The OD is tested by MTT method and the cell apoptosis was detected by flow cytometer with Annexin-V and PI. We found that CD40-ligand would prohibite M231 and ECV cells and could induce these cells apoptosis and INFgama could accelerate the effect. Otherwise,ADM could induce these cells died. Conclusion,CD40-ligand can prohibit both M231 and ECV cells. It means CD40-ligand may hold duplex effect for breast cancer,because CD40-ligand not only prohibit the breast cancer cell but also prohibit the neovascularization of breast cancer. It may be one of the most effective treatment to breast cancer.The second part:We detect CD40 of 10 benign breast mass and 20infiltrating breast duct carcinoma. We examine the Fas and Bcl-2 of these 30 paraffin section using S-P method. We found CD40 is associated with mass size,metastasis of axillary lymph nodes,adherence with local skin. It is important that CD40 is associated with Fas. Conclusion:CD40-ligand should been one effective treatment to the breast cancer patients and the possible mechanism is induced breast cancer cells apoptosis by Fas.
