1. To investigate .the role of TGFpl and sex hormones in the follicular fluid on the day of ovum pick-up during ovulation stimulation cycles; and the role of superphysiological level reproductive hormones on the oocyte maturation, fertilization and development potential.2. To investigate the expression characters of TGFpl and integrinps in endometrial of oocyte donor cycles and normal menstruation cycles, and pinopodes formation in endometrial as consult marker of studying, to analyze the influence of controlled ovarian hyperstimulation on endometrial receptivity.Materials & Methods:1. Follicular fluids (FF) and their matched oocytes were obtained from follicles of 90 women undergoing ovulation stimulation for IVF treatment. TGF betal, E2, P, and LH concentrations in follicular fluid samples that had been collected during transvaginal oocyte retrieval were measured using enzyme-linked immunosorbent assay (ELISA) and enzyme immunoassay (ElA). The maturity, fertilization and embryo quality of the corresponding oocyte were followed. Clinical pregnancies were confirmed by ultrasound at 4 weeks after embryo transfer.2. The endometrial samples were obtained from twenty-five oocytes donors who received ovarian stimulation for IVF-ET. The day of oocyte retrieval was assigned as day 14 in COH cycles. Fourteen healthy fertile women served as a general control group, the urine LHsurge was detection using a commercially available urine kit and the day of LH peak day was designated day 13. Endometrial biopsies were performed every 24-48h, on day!8~23. every sample was divided into two parts, one part examined by immunohistocheniistry for the expression of TGFpland integrinpS, the other by scanning electron microscopy for the detection of pinopodes. Results:1. The mature oocytes group TGFbetal, P and LH concentrations were significantly higher compared to immature oocytes group (2824.4±1414.4pg/ml, 3837.1±942.8 ng/ml and 0.71 ±0.35 miu/ml versus 2015.0±1191.7pg/ml, 3188.1 ±617.4ng/ml and 0.54±0.21 miu/ml respectively, P<0.05). The mean concentration of TGF B 1, E2 and P in the fertilized group ware higher than that in the non-fertilized group (2845.7±1476.7 pg/ml, 3832.0±995.2 ng/ml and 0.73±0.35 miu/ml verses 2044.4 ± 1111.1 pg/ml, 3401.6±607.6 ng/ml and 0.53 ±0.21 miu/ml respectively, P<0.05). The mean concentrations of TGF P 1 and LH in FF with pregnancy group was significantly higher than that in the non-pregnancy group (3631.4 ±1426.3 pg/ml and 0.74±0.25miu/ml versus 2189.2 ±1180.4 pg/ml and 0.52±0.29miu/ml respectively, P<0.05). No significant difference about concentrations of E2 in the follicular fluid which were found in all the groups.2. TGFpl and E2 concentrations in follicular fluid weren't correlated in all groups, Pearson's correlation coefficient r=0.16, P>0.05; TGFpl and LH concentrations in follicular fluid were positive correlated( r=0.26, P<0.05); Progesterone and TGFpl concentrations in follicular fluid were negative correlation (r= - 0.20 .PO.05)..3. The positive TGFP1 and ingetrinps immunostaining in the apical part of the luminal and glandular epithelium was higher in day 18-20(18.84 ± 0.79 ) of COH cycles, TGFP1 and ingetrinp3 less intense immunoreactivity in day 21-23 (21. 88±0.84) of COH cycles. Incontrast, in day 21-23 ( 22.03±0.81) of the natural menstrual cycles, TGF B1 and ingetrinB3 immunostaining of endometrial epithelivun cell were higher intense. The peak mean positive unite of TGFB1 and ingetrinB3 immunostaining of endometrial epithelium cell were 48.44 ±6.75 and 47.92 ±7.01 in COH cycles versus 54.66 ±6.53 and 55.4 ± 6.87 in normal cycles, no significant difference between the two cycles(P>0.05). The fully developed pinopoeds were observed on day 19.3 ± 1.1 in COH cycles, on day 21.0 ±0.8 in normal cycles.4.
