| Objective: To investigate the changes of cytokine TNF-α and IL-6 and organ damages of the liver, lung and kidney in the process of systemic inflammatory response syndrome induced by injecting Escherichia coli Serotype 055: B5 peritonitis intraperitonealy. And also to investigate the anti systemic inflamtnatory response syndrome effect of salvia miltiaorrhiza solution and injection ShengMai and the possible mechanism, to provide a theory base for the clinical use of salvia miltiaorrhiza and injection ShengMai.Methods: The systemic inflammatory response syndrome animal model was established by injecting LPS intraperitonealy. Forty Wistar rats were randomly devided into four groups: Group A ,B, C and D which were treated with normal saline (5ml/Kg) plus LPS (Img/Kg) , SMS (5ml/Kg) plus LPS (Img/Kg), injection ShengMai (5ml/Kg) plus LPSCImg/Kg), SMS(2.5inl/Kg) plus injection ShengMai(2.5ml/Kg) and LPS (Img/Kg) respectively All drugs were given intraperitonealy. Twenty four rats (six rats in each group) were killed at the eighth hourafter establishing SIRS model . Blood was withdrawn and liver, lung, kidney were taken as specimens. LWBC, RBC and PLT were detected with cytometry instrument. 2. TNF-α and IL-6 were determined by double antibody sandwich enzyme linked immuno-sorbent assay . 3. Histological examination was performed to observe the organ injury of liver, lung and kidney. Forty eight hour survival rate was observed in sixteen rats (four rats in each group).Results: l.The changes of WBC. The number of WBC count was 16.43±3.36*109/L in Group A, while it was 6.85±3.38xl09/L 6.57±3.16xl09/L, 6.38±2.68xl09/L in Group B, C and D respectively. The differences between A and B, A and C, A and D were significant, (P<0.05) . But there were no significant statistics changes between Group B, C and D (P>0.05) . RBC and PLT were no significant changes in GroupA, B, C and D (P>0.05). Comparing with TNF-a in Group A (837±104 pg/ml), the number of TNF-α in Group B (237±93 pg/ml), in Group C (364±68 pg/ml ), in Group HD (459±82 pg/ml ) were significantly decreased (P<0.05) .The TNF-α in Group B (237±93 pg/ml) decreased very significantly when comparing with Group A, C and D, (P<0.01) . IL-6 in Group B (307±88 pg/ml), Group C (439±155 pg/ml), and in Group D (367±50 pg/ml) were also significantly decreased as comparing with that of Group A (1224±236 pg/ml) , (P<0.05) . But there was no significant difference among Group B, C and D. 2.The histologicaldamages of the liver, lung and kidney attenuated obviously in Group B, C and D as compared with that in Group A. 3. Forty eight hour survival rate Two rats died in Group A, and the other two survived ,( 50% survival rate ); all rats survived in Group B, C and D, ( 100% survival rate ), There was significant statistics change a± comparing with that in Group A (P<0.05).Conclusions: l.The systemic inflammatory response syndrome rat model induced by injecting Escherictiia coli Serotype 055: B5 peritonitis intraperitonealy is steady, and its procedure was easily performed. 2. Salvia miltiaorrhiza and injection ShengMai can decrease the level of serum TNF-α and IL-6 in the process of LPS-stimulated systemic inflammatory response syndrome, down-regulate the too high inflammatory response, attenuate organ damages of the liver, lung and kidney and increase forty eight hour survival rate obviously. |
