Object: To examine the correlation between the estrogen level in blood serum(E2) the expression of estrogen receptor (ER)and estrogen receptor gene expression of hyperplasia of mammary in stagnation Liver-qi type. Methods: The level of E2 was evaluated by Progesrone Serozyme (EIMA),the expression of ER was tested by Immunohistocheminal assay(S-P),ER gene expression was tested by RT-PCR in 35 cases with mammary hyperplasia of stagnation of Liver-qi type, 15 cases with mammary hyperplasia of disorder of thoroughfare vessel and conception Vessel type and 30 cases of healthy people were taken as control group. Results: (1)serum estrogen level in luteal phase in stagnation Liver-qi type was higher than that in healthy control group significantly (P<0.05), the expression of estrogen receptor(ER) and estrogen receptor mRNA(ERmRNA) expression increased significantly(respectively p<0.05,p<0.05). (2)Compared E2 level in stagnation of Liver-qi type with that in disorder of thoroughfare vessel and conception Vessel type, the former was higher significantly (p<0.01),but the expression of ER and ERmRNA in stagnation of Liver-qi type was lower than that in disorder of thoroughfare vessel and conception Vessel type siganificantly (respectively p<0.05,p<0.05).(3)the positive rate of ER expression was 30 percent and that of ERmRNA expression was 36 percent of hyperplastic tissue, the results were not equal to each other.(4)ER expression of lesion tissues varied with hyperplasia level ;the higher hyperplasia level,the more ER positivecells.(5)ER expression correlated with age,ER positive expression of lesion tissues increased with age. Conclusions:(1)E2 level increase in luteal phase may be an important cause of mammary hyperplasia in stagnation of Liver-qi type. (2)the pathogenesy of mammary hyperplasia disease in different TCM type may not be same absolutely. It indicates: The expression level of E2, ER, ERmRNA can be regarded as microcosmical and objective indexes of syndrome differentiation in Clinic.(3) The cause of ER positive expression disaccord with ERmRNA may be that: RT-PCR is highly sensitive,or ER gene mutate and express nonfunctional protein. (4) ER positive expression of mammary hyperplastic tissue increase with hyperplasia level and patient's age .
