Objective: Disscusing the important role of actin polymerization in the human calcium ionophore A23187-induced acrosome reaction and mechanism of that.Methods: Human spermatozoa are previously dealed with differentconcentration Phalloidin, and then overview the affection of Phalloidin on the human spermatozoa spontaneous acrosome reaction and motility. In a streptolysin (SLO)-permeabilized human sperm model, the spermatozoa are previously incubated in medium with or without Phalloidin for 30 minutes, and calcium ionophore A23187 was added and incubation continued for 15 minutes to induce the acrosome reaction, then we can access the rate of the human acrosome reaction. Finally, we analysis the data and disscuse the role of actin polymerization in the human calcium ionophoreA23187-induced acrosome reaction compared with blank contrast.Results: The differences of human spermatozoa motility and the rate ofspontaneous acrosome reaction between various concentration Phalloidin have no significance(P>0.05). In a streptolysin O(SLO)-permeabilized human sperm model, the difference of the human spermatozoa acrosome reaction between the two groups with or without Phalloidin has obvious significance(P<0.01).Conclusions: Calcium ionophore A23187 can induced the human spermatozoaacrosome reaction. The Phalloidin in the concentration adopted in this experience has no affection on the sperm motility and its spontaneous acrosome reaction , it can work slightly on the acrosome reaction of intact human spermatozoa. In a streptolysin O(SLO)-permeabilized human sperm model, Phalloidin can obviously decreased the rate of human spermatozoa acrosome reaction. The streptolysin O(SLO) can extremely improve the penetrability of spermatozoa outside acrosome membrane and extranuclear membrane, while it don't impact on spermatozoa funcution. The Polymerization of actin play an important role in the course of human spermatozoa acrosome reaction.
