ObjectiveThrough observing th effects of compound ketamine oral solution (CKOS)on rats gastric emptying rate , intestinal transit rate, plasma motilin levels , gastric NMDAR1 and GABAAR - mRNA expression, to find out the effects of CKOS on gastrointestinal motility ,then to evaluate the security of CKOS clinic appliance.Methods1. Effects of CKOS on the rate of gastric emptying , intestinal transit , the level of plasma motilin1) grouping: One hundred and twenty five Wistar rats were randomly divided into five groups. Each group have 25 rats. Then each group was divided into five time points. Each time point have five rats.C group:distilled water 1 ml was irrigated into stomach;T group: sauce solution 1 ml was irrigated into stomach,MID group: MID solution 1 ml was irrigated into stomach, ( Midazolam MID 10mg/kg) ;KET group: KET solution 1 ml was irrigated into stomach, ( Ketamine KET 100 mg/kg);CKOS 组:CKOS solution 1 ml was irrigated into stomach( KET 100 mg/kg, MID10mg/kg and sauce).2 ) Observation and examinationAfter gastric irrigation, 0.2% phenolphthalein 1ml was given . Then differ-ently at 0,30,60,120,360min the rats heads were cut off and 2ml blood was taken. The rats in 30min time point group were take off stomach and small intestine to observe the rate of pigement moving on intestinal and to examine the percentage of pigment in stomach with spectrophotometer.2. The effects of CKOS on gastric NMDAR1 and GABAAR - mRNA expression1) Grouping Thirty male healthy Sprague - Dawley randomly divided into 10 time points. Each time point had 3 rats . Rats in 0 min time point were irrigated with 0. 9% NS 2ul/g for contrast. Others CKOS 2ul/g ( eaquals to KET 100mg/kg,MID 10mg/kg).2 ) fixation of gastric tissueAfter drug administration respectively at the time of 0,5,10,15,30,60,90, 120,240,360min, catheter was inserted from right heart to irrigate 0.9% cold Na Cl,the colour of liver changed from red to white (150ml) , extra fluid are removed from venecava. Then after all body organs were fixed with 4% polymalde-hyde stomach were taken out and put into 4℃ refrigerator.3 ) Observation and examineObserve the changes of NMDAR1, GABAA R - mRNA in rats stomach by Immune histochemistry staining and in situ hybridization . Microscpoe ( flu-roscense) image analyzing system was used to analyze brain sections.Results1. Effects of CKOS on the rate of gastric emptying and intestinal transit ,the level of plasma motilinThe motilin levels in C group at 30,60 min were higher than 0 min time point(P <0. 01). The motilin levels in T group at 30,60min were also highe than 0 min time point and coresponding points in C group ( P <0.01). The motilin levels at 30,60min in KET, MID,CKOS groups was less than coresponding points in C group. There was not difference between KET, MID,CKOS groups (P > 0.05 ).
