Studies On Preparation Process And Quality Standard Of Zhiyang Lotion

Objective: To ascertain the extraction process of the Chinese herbal preparation Zhiyang Lotion , and to establish quality standard of it.Methods: The preparation proess was divided into two parts. 1,By orthogonal design ,with extractive yield of volatile oil as an index, the optimal oil extraction condition was studied. Three factors were selected including water consumption, soaking time and extracting time. 2, By orthogonal design , with the content of Matrine determined by HPLC as a quantitative index, the optimal water extracting condition was studied. Four factors were selected including water consumption, extracting times , extracting time and the concentration of alcohol-sediment.TLC was used in qualitative identification for Zhiyang Lotion. Cnidium monnieri (L) Cusson, Sophora flavescens Ait, Zanthoxylum bungeanum Maxim, Stemona tuberosa Lour were identified by TLC using 0. 5% CMCNa-silica gel G with dots clear and reproducible. Cnidium monnieri (L) Cusson was identified with benzene-ethyl acetate (30 : 1) as developer and observed at 365nm using ultraviolet lamp. Sophora flavescens Ait was identified with benzene-acetone-methanol (8 : 3 : 0.5) and the upper mixture of toluene-ethyl acetate - methanol-distilled water (2 : 4 : 2 : 1)as developer and bismuth potassium iodide -acohol (2 : 1) as coloration . Zanthoxylum bungeanum Maxim was identified with petroleum ether (60 90℃ )-ethyl acetate (19 ∶ 1) as developer and dinitrophenylhydrozine solution as coloration. Stemona tuberosa Lour was identified with benzene-acetone-methanol (8 ∶ 3 ∶ 0. 5) as developer and bismuth potassium iodide -acohol (2 ∶ 1) as coloration.Matrine in Zhiyang lotion was measured by HPLC as a quantitative index.
