Objective: To observe the effects on praxiological characters and blood lipid and cerebral microcirculation and pathological change of brain with the apolipoprotein E gene knock-out mice. Methods: Different weeks-old ApoEKO mice were devided into youth model group, adult model group and eldly model group randomly. Normal C57BL/6J mice were devided into youth normal group and eldly normal group. With the methods of behavior, We investigated the ability of learning and memory and the ability of moving in control and the ability of resisting the tiredness in ApoEKO mice. With the same model group and youth normal group, We observed the cerebral microcirculation, blood lipid and pathological changes of brain tissue. Results: During the praxiological test, the ability of learning and memory of eldly ApoEKO mice decreased more than the same weeks-old C57BL/6J mice. With the age adding up, the ability of learning and memory of ApoEKO mice decreased. There was no difference among the mice at the ability of moving in control. The ability of resisting the tiredness of ApoEKO mice reduced more than the same weeks-old C57BL/6J mice. With the age adding up, the ability of resisting the tiredness of ApoEKO mice reduced. We also found that the total cholesterol (TO ,low density lipprotein cholesterol (LDL-C) and high density lipprotein cholesterol (HDL-C) improved obviously in ApoEKO mice. Meanwhile the cerebral microcirculation reduced dramatically in ApoEKO mice. The pathological changes of brain tissue included neurofibrillary tangles, senileplaque, neuron loss and cerebral amyloid angiopathy. Conclusions: From the above all we can conclude that apolipoprotein E gene knockout could decrease the ability of learning and memory and resisting the tiredness, improve the level of blood lipid, reduce the level of cerebral microcirculation and bring about the pathological changes of brain...
