Objective: To reproduct a rat model of Liver-Yang Hyperactivity Syndrome with hypertension and grope for the mechanism of Tianma Gouteng decoction curing it, the compound prescription which restrain the hyperactivity of the liver yang.Methods: To reproduct a rat model of Liver-Yang Hyperactivity Syndrome with hypertension when spontaneously hypertensive rats (SHR) are used, administrated with Fuzi decoction and treated by Tianma Gouteng decoction. The rat syndrome is observed with rank score. The rats' blood pressure is measured from caudal artery by electronic sphygmomanometer. The plasma level of angiotensin II (Angll) and aldosterone (ALD) are measured by radioimmunoassay (RIA). The change of proteome of rat liver is analyzed by two-dimensional gel electrophoresis (2-DE).Results: â‘ After drenching Fuzi decoction for six weeks, the rats appear such symptoms: the puffed and lackluster hair, loss of appetite, an increase in saliva and appetite, conjunctival congestion, irritation, increase in water intake and red tongue with little coating. Those are manifestation of Liver-Yang Hyperactivity Syndrome with hypertension. â‘¡After treatment of highdose or lowdose Tianma Gouteng decoction, the symptoms of rats are as following: the compact and lustrousness hair, conjunctival congestion lessen, irritation take a turn for better, decrease in water intake, fall in blood pressure. The plasma level of Angll falls sharply in highdose group of Tianma Gouteng decoction. And it is observed that there is no effect on the plasma level of ALD to the treatment of Tianma Gouteng decoction. â‘¢Analysis of the proteome of rat liver: The electropherograms of rat liver protein are similarin the groups of spontaneously hypertensive rats (SHR for short), Liver-Yang Hyperactivity Syndrome (GYSK for short) and Tianma Gouteng decoction (TMG for short). A majority of proteins distribute in PI 5-8. There are 716+58 protein spots in the 2-DE map of the SHR group and 305 spots are matched to all members in the group, 875 + 19 protein spots in the GYSK group and 409 spots matched to all members in the group, 899 + 25 protein spots in the TMG group and 461 spots matched to all members in the group. There were only 13 protein spots examined in SHR group and 27 protein spots examined in GYSK group by comparing the two groups. And 28 differentianlly expressed protein spots (>5 fold) have been found in the two groups, 15 of them down regulate and 13 of them up regulate in quantity. There were only 44 protein spots examined in TMG group and no protein spot was examined in GYSK group by comparing the two groups. And 29 differentianlly expressed protein spots (>5 fold) have been found in the two groups, 14 of them down regulate and 15 of them up regulate in quantity.Conclusions: Using SHR plus drenching Fuzi decoction, we can reproduct an ideal Chinese syndrome model: a rat model of Liver-Yang Hyperactivity Syndrome with hypertension. Tianma Gouteng decoction can lessen the symptoms, fall blood pressure and fall the plasma level of Angll of the model. Influenting the plasma level of Angll may be one of the mechanisims of Tianma Goteng decoction reducing blood pressure. The proteins of rat liver differentianlly expressed indicate that some protein spots may be associated with Liver-Yang Hyperactivity Syndrome with hypertension and the target of Tianma Gouteng decoction on the syndrome. These results set bases for further research. |
