Objective To explore the antitumor effects and the mechanism of GFW on the S180 bearing tumor mice . The main study Point is the effect of GFW on the expression of PCNA ,Survivin , p21waf/cip and T lymphocyte sub-group . Method The S180 mice sarooma model was used to detective the inhibiting effects of GFW , observed the ultrastructural change by electron microscopy. The mRNA transcription of Survivin and the protein expression of the proliferating cell nuclear antigen(PCNA)and p21waf/cip were investigated by using in situ hybridization(ISH) and SP immunohistochemical techniques respectively. Flow cytomertric analysis was used to determine the expression in T lymphocyte sub-group .Result For S180 sarcoma , the inhibiting ratio of GFW is 34.11% . The positive protein expression of PCNA was markedly declined in the GFW group when compared with the model group (p <0.01) . The suppress cancer gene p21waf/cip was contrary to PCNA , its protein expression showed to rise in the GFW group , there was significant difference between GFW group and model group (p<0.01). Some typical apoptotic cells and apoptotic bodies were observed through electron microscope . The positive expression of Survivin mRNA was decreased in the GFW group when compared with the model group (p<0.01) respectively. Our study also showed the GFW could regulate the expression of CD8+ and the rate of CD4 / CD8 approaching normal . Conclusion GFW has the antitumor function and can adjust immune . It could decreased the expression of Survivin mRNA and the expressionprotein levels of PCNA, rised the expression protein of p21waf/cip. GFW regulated the level of T cell subgroup approaching normal .
