The Research On CpG ODN's Immune Enhancement When Co-administrated With Recombinant HBsAg Vaccine

Objective In some in vivo and in vitro experiments,it has been confirmed that the unmethylated CpG dinucleotides in bacterial DNA or synthetic oligodeoxynucleotides(ODN) has the ability to induce B cell proliferation and Ig secretion,monocyte cytokine secretion,and activation of NK cell lytic activity and INF-γsecretion. The potent immune activity by CpG ODN suggests possible utility for enhancing immune responses to vaccine. To a certain degree,the HBsAg recombinant vaccine which used universally has some initiative in preventing the infection of HBV.But through some statistics estimation ,it is demonstrated that nearly 10～15% among the vaccinated people have not produced ideal acquired immunity. Thus,in order to overcome the defect ,many researchers are attempting to develop a new vaccine in which particular adjuvant is added.Under these circumstances,this experiment is trying to inspect the influence on the C57BL/6 mice coming from the CpG ODN,through HBsAg recombinant protein vaccine co-vaccinating with the unmethylated CpG ODN,which is even been pondered whether the CpG ODN can be used in the production of vaccine as a new adjuvant,stimulating immune system to produce effective responses.Method The C57BL/6 mice were put into three groups,namely,contrast group,vaccine group,and the group containing vaccine adding CpG ODN.The mice in contrast group were inoculated with N.S;the second group were with HBsAg recombinant protein vaccine lonely;and the mice in the third one were vaccinated with vaccine adding CpG ODN .The vaccination schedule was 1st,15th,22nd.The spleen suspension were harvested 29 days later after the inoculation .Four cytolytic immunological tests were done through MTT essay,including HBsAg specific stimulated CTL,NK none-antigen specific CTL test,and ConA specific lymphocyte transforming test;under the ELISA assay,cytokines INF-γ,IL-12(P70),IL-4 and HBsAb in serum were...
