| Objective: In search of a new and more effective sort of herb product for protecting gastric mucosa lesions induced by NSAIDs(Non Steroidal Anti-inflammatory Drugs), we studied the role of JianWei Mixture in preventing NSAIDs-induced gastric lesions by analyzing histomorphology, mucosal blood flow, 6-keto-PGF1α, TXB2 and ET-1 within rat models. On the basis of this study, the effect of JianWei Mixture to relieve NSAIDs-induced gastric lesions has been estimated, and the mechanism of protective function is explored as well. Methods:1.Animal categorization: In this experimentation, 60 SD rats were randomly divided into 6 groups that contained 10 rats each. 2.Establishment of animal models: All these 6 groups were designed for different purposes. Briefly, Group 1 (negative control group),normal saline 2ml/100g; Group 2 (modeling control group) ,normal saline2ml/100g either; Group 3 (positive control group), Sucralfate solution 2ml/100g; Group 4 (low does group of JianWei Mixture), Jian Wei Mixture 1ml/100g; Group 5 (medium does group of JianWei Mixture),JianWei Mixture 1.5ml/100g; Group 6 (high does group of JianWei Mixture), Jian Wei Mixture 2ml/100g. In seven days the rats were orally administered with relative drugs once time for each day, The rats of Group 2-6 accepted oral administration of Asprin (100mg/kg) in 30 minutes. Animals were withheld oral food for 24hrs and water for 12hrs before last oral administration.3.Observation items: measurement of mucosal blood flow; estimation for changes of gastric mucosa by endoscope and microscope; quantitative analysis for expression of 6-keto-PGF1α, TXB2 and ET-1.Results: There were significant improvements for NSAIDs-induced lesions in positive control and JianWei Mixture groups in comparison to modeling groups. GMBF, 6-keto-PGF1α, TXB2 and expressions were significantly increased in positive control and JianWei Mixture groups. Moreover, ET-1 expression was significantly decreased in JianWei Mixture groups only.Conclusion: JianWei Mixture can significantly improve NSAIDs induced gastric mucosal lesions by increasing expression of GMBF,6-keto-PGF1α,TXB2, and decreasing expression of ET-1. Therefore, it is evident that JianWei Mixture can protect gastric mucosa through multiple approaches. |
