| ObjectiveThe effect of zinc and zinc complex on inhibiting psoriasis development in mouse vaginal epithelium model and mouse tail scale epidermis model were explored. Methods1. Effects of zinc and zinc complex on mouse vaginal epithelium modelFemale Kunming mice were used to established mouse vaginal epithelium at estrus model .They were randomly divided into nine groups: negative control, positive control, low, middle, high dosage of ZnCl2, low , middle ,high dosage of Zn-EDTA and zinc gluconate. The mice were killed after the last administration, and the vaginal mitosis index was observed under optical microscope.2. Effects on mouse tail scale epidermis modelsMale mice were randomly divided into nine groups indicated as above. The number of scale with granular layer per 100 scales were examined by opticalmicroscope.3. Effects on removing other essential elementsTo investigate zinc, copper, iron, magnesium, manganum and calcium contents, cardiac, liver, spleen, lung, kidney, tail, brain and muscle of mice in all groups were taken and digested to analyze the elements concentration by atomic absorption spectrophotometry.4. Effects on recovering the anti-oxidizing systemThe activities of SOD, GSH-PX and the content of MDA in the homogenate of liver, kidney of the mice were determined in each group.5. Effects on the serum level of IL-2 and IL-10Radioimmunoassay was applied to measure the serum level of IL-2 and IL-10 in each group. Results 1. Effects of zinc and zinc complex on mouse vaginal epithelium modelEvery zinc spiked groups as well as methotrexate group could obviously inhibit the mitosis of vaginal epithelial cells (P<0.05). No statistical significance was found between each zinc spiked group and the positive control group (P>0.05).2. Effects on mouse tail scale epidermis modelsThere was statistical significance between each zinc spiked group as well as the positive control and the negative control group (P<0.05).3. Effects on removing other essential elementsZinc contents of each zinc spiked group in tails were higher than that of the negative control group and methotrexate group (P<0.05). No statistical significance was found between the negative control group and methotrexate group (P>0.05). In comparison with zinc contents, no statistical significance existed among the groups in the cardiac, liver, spleen, lung ,kidney, tail, brain and muscle (P>0.05).In comparison with copper, iron, magnesium, manganum, and calcium contents, no statistical significance existed among the groups in the cardiac, liver, spleen, lung, kidney, tail, brains and muscle (P>0.05).4. Effects on recovering the anti-oxidizing systemIn comparison with the corresponding data of the negative control group (P<0.05), statistical significance existed in every zinc replenished group and methotrexate group in terms of the activities of SOD, GSH-PX and the content of MDA in livers, while no statistical significance existed in kidneys.5. Effects on the serum level of IL-2 and IL-10The serum level of IL-2 in each zinc spiked group as well as methotrexate group was obviously lower than that in the negative control group (P<0.05).The serum level of IL-10 in each zinc spiked group as well as the negative control group was obviously lower than that in methotrexate group (P<0.05). The serum level of IL-10 increased in each zinc spiked group , but it was no statistical significance(P>0.05). Conclusion 1. As a result of zinc replenishment, the mitosis of vaginal epithelial cells was obviously inhibited, and the formation of granular layer in mice tail scale was significantly promoted.2. Zinc replenishment could obviously enhance zinc contents in tails and had no obvious effect on other essential elements such as copper, iron, magnesium, manganum, and calcium.3. It could also enhance the activity of the antioxidant enzyme and reduce peroxidization of lipid.4. It could significantly lower the serum level of IL-2 and has no obvious effect on that of IL-10. |
