OBJECTIVETo study the effect of N-acetylcysteine (NAC) in the pathgenesis of schistosomiasis liver pathological changesMETHODS1. Murine schistosomal hepatic pathological changes model was built by making the mice transcutaneous infected with S. japaonicum cercariae.2. Models were treated with NAC by intragastric way in different time and different dosage.3. Histochemical stainingThe mice were killed at the end of experience, their liver were cut off and embedded with 4% paraform, then the pathological changes and the deposition of collagen in liver were observed by HE and VG staining.4. The number, area of single egg granuloma and areas of collagen fibroses were measured with Image Analysis Instrument, the levels of hydroxyproline (Hyp) were detected.5. The indexes about hepatic pathological changes in serum and liver tissues were detected: inducible Nitric oxide synthase (iNOS), Nitric Oxide (NO), Glutathione peroxidase (GSH-PX), Reduced glutathione hormone (GSH), Malondialdehyde (MDA), Superoxide dismutase (SOD).6. Statistics analyses were perfomed with software SPSS 11.5 ANOVA. RESULTS1. The liver was damaged, and the numbers of the egg granuloma and the deposition of collagen increased after infection. Variety acute egg granuloma and a lot of inflammatory cells were observered in liver tissue. The hepatic SOD, GSH concentration were lower, The concentrations NO, Hyp and MDA and the activities of iNOS of the hepatic tissues were all higher.2. The liver damage and fibrosis degree were reduced after using NAC. The number and areas of acute egg granuloma in preventive group or treatment group were decreased obviously than those in infective control group. The concentration of Hyp, NO, iNOS and MDA in liver and GSH, MDA in serum were reduced obviously. The level of SOD, GSH in liver and GSH-PX, SOD in serum were increased resepectively.3. The number and area of egg granuloma were affected by the time and dosage of using NAC. The effect of reducing number and area of egg granuloma in preventive group (using NAC as soon as infection) was better than those in treatment group (using NAC after infection 42 days). The effect reducing number and area of egg granuloma in 200ml/kg NAC was better than those in 400ml/kg NAC. The number and ares of "+" level single egg granuloma in preventive a group were smaller than that in treatment a group.4. The numbers of the egg granuloma and the deposition of collagen in the prevented a group were fewest for all groups. In the group, the hepatic Hyp, MDA, iNOS, NO concentration were lower, but SOD was higher than that in infected group.5. NAC may make the hepatic Hyp, MDA, iNOS and NO concentration tend to the physiological state of the mice, which relate to the effects with its dose.CONCLUSIONNAC has the effects on inhibiting increase or promoting decrease of the liver tissue pathologic changes and hepatic fibroses. The effect may associate with the decrease of the hepatic Hyp, NO and MDA concentration and the tissue iNOS activity, with the increace of the hepatic SOD. These effects were related with NAC dosage. These effects were better in preventive group than in treatment group.
