Currently, traditional Chinese medicine has been extensively applied in clinical, whereas its toxicity, especially genetic toxicity, is not clearly elucidated. In the present study, the DNA damage of mouse lymphocytes induced by herbs for regulating QI flow and their compatibilities, which were frequently used clinically, were investigated with the single cell gel electrophoresis (SCGE) technique and micronucleus test.(1) Using SCGE method, we studied the DNA damage of mouse lymphocytes induced by single herbs for regulating QI flow. The Kuming mice were treated with 1/3×clinical equivalent dose, clinical equivalent dose and 3×clinical equivalent dose by intragastric administration for 1d, 3d, and 7d, respectively. The results showed here that Citrus trticulata Blance, Allium macrostemon Bge, Cyperus rotundus L., Citrus aurantium L, Citrus medica L.var.satcodactylis Swingle, Litchi chinensis Sonn, Rosa rugosa Thunb, Citrus reticulata Blanco and Citrus grandis (L.) osbeck had no influence on peripheral blood lymphocyte cell DNA in mice. However, The lymphocytes DNA were damaged by Diospyros kaki Thunb, Aquilaria sinensis (Lour.) Gilg, Melia toosendan Sieb.et Zucc, Aspongonpus chinensis Dallas, Areca catechu L, Aucklandia lappa Decne, Lindera aggregata (Sims) Kost, Nardostachys chinensis Batal and Canavalia gladiata (Jacq) DC at different dose and time.(2) Using SCGE method, we also studied the DNA damage of mouse lymphocytes induced by the compatibilities of herbs for regulating QI flow. The results showed that itrus reticulata Blance, Citrus aurantium L, and Allium macrostemon Bge. could reduce the toxicity of their compatibilities to some extent including Pinellia ternate (Thunb.) Breit, Magnolia officinalis Rehd.et Wils and richosanthes kirilowii Maxim., In addition, we found that Aquilaria sinensis (Lour.) Gilg and Aucklandia lappa Decne could degrade each other's toxicity through interaction, and Corydalis yanhusuo W.T.wang could lower part of the toxicity of Melia toosendan Sieb.et Zucc after short-term administration.(3) Investigation of the compatibilities of Citrus reticulata Blance and Pinellia ternate (Thunb.) Breit effect over DNA of peripheral-blood lymphocyte in mice by SCGE and micronucleus test shows that: the damage effect of Pinellia ternate (Thunb.) Breit works 1h after administration, however, the damage effect can not be detected 24h after administration.The results indicate that our organism can repair the DNA damage caused by Pinellia ternate (Thunb.) Breit ,it also indicate that SCGE can detect the lower and the earlier DNA damage and it is more sensitive than micronucleus test.
